The invention discloses
bacillus subtilis chassis cells for
vitamin B6 production and a construction method of the
bacillus subtilis chassis cells, and belongs to the technical field of bioengineering. The
chassis cell is obtained by performing systematic knockout on eight genes including upp, srf, spo, aprE, nprE, nprB, mpr and wprA in
bacillus subtilis 168 through a
gene engineering technology, wherein the eight genes include the upp
gene, the srf
gene, the spo gene, the aprE gene, the nprE gene, the nprB gene, the mpr gene and the wprA gene. Precise knockout of the gene is realized by adopting a
homologous recombination method, and the obtained chassis
cell shows remarkably optimized physiological characteristics: the growth rate and the glucose
metabolism rate are remarkably improved, and meanwhile, the sporulation rate and the
extracellular protease activity are remarkably reduced. Experimental results show that the modified chassis
cell has excellent suitability with a
heterologous gene element of a
vitamin B6 synthesis pathway, and can efficiently synthesize
vitamin B6. Therefore, the recombinant bacillus subtilis chassis cell provided by the invention has important application value in the field of microbiological manufacturing of
vitamin B6.
