The invention discloses an in-vitro culture method for the pearl shell mantle tissue and a special culture solution thereof. The culture medium comprises hydrolyzed pearl liquor, shell liquor, antibiotics and seawater. The method conventionally comprises the following steps: taking the material from a nacre to obtain the mantle epithelium, removing the skirt band part, cutting the tissue into tissue blocks with the size of about 1mm to 2mm after purification treatment, putting the tissue blocks in the protease solution for digestion, and adding the culture solution to stop the digestion. The preliminarily digested tissue blocks are respectively put into a culture dish or cling to the pearl core which is covered by adherence substance. The cultured pearl shell mantle tissue grows in good conditions; after 4 hours, the cells are emigrated from the tissue blocks; and after 2 days in-vitro pearl sacs are formed effectively. The method can be used to inject the anucleate pearl sacs at different positions in the pearl shell with the inserted pearl nucleus so as to culture the anucleate pearl. The method has low cost, a high survival rate, a high pearl formation rate, and little damage to the nacre.

The invention provides a method of culturing pearls by using pearl nucleuses with tissue engineering scaffolds. The method is characterized in that a layer of degradable high-molecular polymer with a uniform thickness, serving as a tissue engineering scaffold, is attached to the surface of a pearl nucleus which is to be implanted into a pearl mussel; the tissue engineering scaffold is three-dimensional network space capable of accommodating and growing cells; mantle tissue of pearl shells is separated into single existent free mantle cells by using the enzyme digestion method and a mantle cell suspension with a certain cell density is prepared; mantle cells are attached to the pearl nucleus and then the pearl nucleus and the mantle cell suspension are implanted in a nucleus insertion position of the pearl mussel for culture of a nucleus pearl. The method is simple in operation, requires no in-vitro culture of cells and solves the problem of high technology and equipment requirements of a cell culture method; aseptically packaged pearl nucleuses can be directly transported to culture sites for operation and the transportation is convenient; the method has low requirements for equipment and operators, reduces the production cost greatly and is suitable for large-scale production and application.

By the method for culturing the nucleated pearls, the nucleus implanting and keeping rate can be effectively improved by more than 90 percent, and spherical, heteromorphic and image nucleated pearls with a magnetic health care effect can be produced.
