A receiving block formed of soft and resiliently flexible material, such as an elastomeric foam, may be impaled on the prongs. The present application relates to the field of biological materials, in particular to a preparation method and application of GMS@EVs microspheres loaded with SNAP-VHH fusion protein. The present application constructs a gelatin microsphere delivery system (GMS@EVs) with capture and slow-release functions. The system specifically and efficiently captures CD9-positive exosomes secreted by mesenchymal stem cells through the stable fixation of SNAP-VHH fusion protein on the surface of gelatin microspheres, and realizes the efficient extraction of CD9-positive exosomes from the culture supernatant of mesenchymal stem cells. The present application provides specific reaction sites for the fixation of SNAP-VHH fusion protein by grafting O6-[4-(aminomethyl)benzyl]guanine on the gelatin microspheres, so that the SNAP-VHH fusion protein is stably loaded on the gelatin microspheres. Through experiments, the GMS@EVs microspheres prepared by the present application have controllable slow-release properties for exosomes, and still maintain significant fluorescence signals 48 hours after subcutaneous injection in nude mice, which is at least 16 times longer than the slow-release time of free exosomes (30 minutes of rapid diffusion), realizing the sustained release of exosomes in the local wound and enhancing the durability of the therapeutic effect.

The method comprises the following steps: (a) culturing a first generation of ovarian cancer transplantation tumor in a first batch of nude mice by a cell suspension subcutaneous injection method; (b) obtaining the first-generation transplanted tumor tissue obtained in the step (a), and preparing the first-generation transplanted tumor tissue into a solid tumor block suitable for transplantation; and (c) transplanting the solid tumor block prepared in the step (b) to the subcutaneous part of a second batch of nude mice to construct a subculture transplantation tumor model. Compared with a traditional cell suspension direct injection method, the adopted subculture solid tumor mass transplantation method can remarkably improve the tumor formation success rate and the tumor growth speed. The model constructed by the method retains more tumor interstitial components, is closer to the biological characteristics of clinical tumors, and provides a more stable and reliable in-vivo experimental platform for drug screening, pathogenesis research and personalized treatment of ovarian cancer.
