A method for identifying and isolating cells which produce secreted proteins. This method is based upon a specific characteristic or the expression level of the secreted protein by transiently capturing the secreted protein on the surface of an individual cell, allowing selection of rare cell clones from a heterogeneous population. Also provided is the use of this method to generate cells which produce a desired level of secreted protein or secreted protein of a particular characteristic(s), and organisms which possess such cells. In particular, the method allows rapid isolation of high expression recombinant antibody-producing cell lines, or may be applied directly to rapid isolation of specific hybridomas, or to the isolation of antibody-producing transgenic animals. Microfluidic device for cell and particle separation

A microfluidic separation device includes a microchannel formed in a substrate and being defined at least by a bottom surface, a first side wall, and second side wall. Fluid containing particles or cells is flowed through the microchannel from an upstream end to a downstream end. The downstream end terminates in a plurality of branch channels. A plurality of vertically-oriented electrodes are disposed on the first wall and on the second wall opposite to the first wall. A voltage source is connected to the plurality of opposing electrodes to drive the electrodes. The opposing, vertically-oriented electrodes may be used to focus a heterogeneous population of particles or cells for subsequent downstream separation via additional electrodes placed on one of the side walls. Alternatively, the opposing, vertically-oriented electrodes may be used to spatially separate a heterogeneous population of particles or cells for later collection in one or more of the branch channels.

The method includes providing a tissue sample comprising fixed cells; exposing the sample to first and second ligands that recognize different marker proteins found at the same cellular location, thereby forming a ligand-labeled sample; exposing the ligand-labeled sample to first and second labeling reagents, the first labeling reagent binding to the first ligand and the second labeling reagent binding to the second ligand, the first and second labeling reagents each forming distinct pigments; and identifying the number of cells that display only one particular pigment, or more than one pigment, by the different coloration of the cellular location labeled by the distinct pigment.

Novel, nanoparticle-based vaccines are provided that elicit an immune response to a broad range of infectious agents, such as influenza viruses. The nanoparticles comprise a heterogeneous population of fusion proteins, each comprising a monomeric subunit of a self-assembly protein, such as ferritin, joined to one or more immunogenic portions of a protein from an infectious agent, such as influenza virus. The fusion proteins self- assemble to form nanoparticles that display a heterogeneous population of immunogenic portions on their surface. When administered to an individual, such nanoparticles elicit an immune response to different strains, types, subtypes and species with in the same taxonomic family. Thus, such nanoparticles can be used to vaccinate an individual against infection by different Types, subtypes and / or strains of infectious agents. Also provided are specific fusion proteins, nucleic acid molecules encoding such fusion proteins and methods of using nanoparticles of the invention to vaccinate individuals.

The invention discloses a sterilization method for edible mushroom culture. Compared with the prior art, the sterilization method for edible mushroom culture has the advantages that heterogeneous population caused by the reason that the compost is not sterilized thoroughly, the inoculating process is not standard, the strains carry disease germs, the site conditions are poor and the like can be thoroughly sterilized, and the edible mushroom is prevented from being infected.
