By optimizing the pH conditions, antibody binding amount, purification and dilution conditions during the preparation of the gold-labeled conjugate pad, as well as the preparation conditions of the T and C lines in the NC membrane, and further employing a pretreatment method adapted to *Ophiopogon japonicus* matrix samples, the resulting kit achieves performance that meets standards and surpasses existing product types in the detection of three plant growth regulators—paclobutrazol, chlorpyrifos, and 6-BA—in *Ophiopogon japonicus* samples. It exhibits high sensitivity, specificity, accuracy, and stability.
