Microfabricated cell sorter

The invention provides a microfabricated device for sorting cells based on a desired characteristic, for example, reporter-labeled cells can be sorted by the presence or level of reporter on the cells. The device includes a chip having a substrate into which is microfabricated at least one analysis unit. Each analysis unit includes a main channel, having a sample inlet channel, typically at one end, and a detection region along a portion of its length. Adjacent and downstream from the detection region, the main channel has a discrimination region or branch point leading to at least two branch channels. The analysis unit may further include additional inlet channels, detection points, branch points, and branch channels as desired. A stream containing cells is passed through the detection region, such that on average one cell occupies the detection region at a given time. The cells can be sorted into an appropriate branch channel based on the presence or amount of a detectable signal such as an optical signal, with or without stimulation, such as exposure to light in order to promote fluorescence.

A cell sorting chip and a cell sorting technology are to be established which can positively detect and sort a specified cell for cell separation and detection using micro flow paths formed on a substrate, whereby a cell analyzing / sorting device is provided which uses an inexpensive disposable chip replaceable for each sample. To this end, micro flow paths formed on the substrate are formed, and cells are roughly sorted in a first stage and then finely sorted in a second stage. More specifically, cells are sorted roughly by using scattered light or according to intensity of luminescence in the first stage. In the second stage, the roughly sorted cells are sorted with high precision using image recognition.

The chip of the invention can effectively and rapidly fix the magnetic beads, increase the sample size of cells, improve the analysis flux, reduce the dead volume of sample introduction and reduce the retention and cross infection.

Methods and apparatus are provided for culturing cells under conditions for determining cellular differentiation and for separating cells from culture media based on differentiation. The apparatus comprises a bioreactor, media reservoir, a magnetic cell separator, an inlet port for adding magnetic particles to the bioreactor, and a circulating pump, wherein the bioreactor, media reservoir, magnetic cell separator and inlet port are on a single fluid circuit. The present method and apparatus provides a method for separating cells from culture without removing the cells from culture, so that un-selected cells may be returned to the bioreactor for further culture. The method employs magnetic labeling in culture, where the magnetic label specifically identifies cells to be distinguished, either by separation or retention in the culture. The method and apparatus are further designed to comprise means for electromechanical stimulation of hum embryonic stem cells for preparation of electrically responsive tissue.

The cell-delivery unit of a high-throughput electrophysiological testing system is implemented as a reusable movable unit suitable for repetitive delivery of cells to a disposable, multi-aperture patch-clamp tray. An electric field emanating from the patch aperture is used to align the dispenser with the aperture. A set of electrodes in the nozzle of the dispenser is used to detect the electric field and effect the alignment. According to another aspect of the invention, dielectrophoretic fields produced by sets of electrodes in the nozzle form a retaining cage that is used first to suspend a test cell directly above the patch aperture and then to urge the cell toward it. Methods for Isolating Monocytes

The present invention provides HIDE1 as novel monocyte markers. Since HIDE1 are membrane proteins, monocytes can be specifically detected by using antibodies that bind to HIDE1. Further, HIDE1-positive monocytes can also be collected from peripheral blood or the like using a cell sorter, magnet, or such. Monocytes that can be prepared based on the present invention are useful in cell immunotherapy.

A 3-dimensional PDMS cell sorter having multiple passages in a PDMS layer that follow the same path in a DEP separation region and that are in fluid communication with each other within that region. The passages may differ in width transverse to the flow direction within the passages. Flat plates may sandwich the PDMS layer; each plate may have a planar electrode used to generate a DEP field within a sample fluid flowed within the passages. The DEP field may concentrate target cells or particulates within one of the passages within the DEP separation region. The passages may diverge after the DEP-separation region, leaving one passage with a high concentration of target cells or particulates. Techniques for manufacturing such structures, as well as other micro-fluidic structures, are also provided.

The invention discloses a method for preparing foot-and-mouth disease virus-resistant (FMDV) transgenic livestock by expressing FMDV-resistant ribonucleic acid interference (RNAi), which comprises the following steps of: linearizing an FMDV-resistant RNAi recombinant Lenti-virus vector which carries a short hairpin RNA (shRNA) base sequence and a green fluorescent protein (GFP) reporter gene and transfecting fetus fibroblasts of normal livestock; sieving transgenic nuclear donor cells by using the GFP reporter gene and a flow cell sorter and introducing nuclear donor nuclei into denucleated oocytes of the livestock to obtain a reconstructed embryo; and immigrating the reconstructed embryo into the uterus of surrogate livestock to obtain an offspring, namely the FMDV-resistant RNAi heterozygote transgenic livestock. Heterozygote transgenic male livestock and heterozygote transgenic female livestock are normally mated or reproduced by artificial insemination so as to obtain homozygote transgenic livestock. The transgenic livestock prepared by the method can express the FMDV-resistant RNAi, obviously reduces the FMDV infection rate and has the capability of resisting foot-and-mouth disease (FMD).
