The application provides a VIGS-based
sida gene silencing
system and method, and belongs to the technical field of
genetic engineering. The application provides a recombinant
gene silencing vector suitable for
sida VIGS, which is based on a BSMV_pCaBS-gamma-LIC carrier as a basic skeleton carrier, wherein a silencing fragment of a
target gene of
sida is connected, and the silencing fragment is transformed into
agrobacterium with a BSMV alpha carrier and a BSMV beta carrier respectively, and then is used to infect
nicotiana benthamiana to proliferate
virus liquid; and then, based on a mechanical friction method, the sida leaf is infected to complete the silencing of the
target gene. The VIGS provided by the application can successfully
silence the
gene, and provides a feasible method for studying the gene function of sida; the method can be implemented in a laboratory, and does not need
in vitro transcription and p2 laboratory extraction, thereby saving a large amount of cost; meanwhile, the yield of
virus particles is improved, and the success rate and efficiency of silencing are improved.
