The invention discloses a modeling method for grading quality of Xinjiang red dates in a near infrared range, which relates to modeling methods for grading quality of natural products in the near infrared range, and solves the problem that the prior randomly selected red date sample near infrared ranges differ greatly, main spectrum characteristic factors influencing spectrum change cannot be separated, and consequently, the grading modeling effect is poor. The modeling method provided by the invention comprises the following steps of: initially choosing appearance characteristics of samples, then, scanning the red date samples of same batch one by one; obtaining average spectrum of the batch of red date samples after performing proper spectrum treatment, and comparing the average spectrum with an original sample spectrum chart to screen the samples according to similarity so as to select the samples with greater spectrum difference, performing spectrum scanning again on the residual samples, and repeatedly adjusting and gradually subdividing the residual samples, until the spectrum difference of the batch of samples is in a needed accuracy range; and encoding the separated characteristic factors to create a sub-model. With the method, the characteristic factors of the Xinjiang red dates are successfully separated, and the sub-model is successfully created, so that the method provided by the invention is suitable for grading the quality of fruits, such as red dates, apples, pears and the like.

The method of preparation of urinary protein includes steps of: 1) performing ultracentrifugation to a urine sample at normal temperature for a certain time and rejecting the supernatant and maintaining a precipitate; 2) adding a proper amount of resuspending buffer liquid to the precipitate in the step 1) to re-suspend the precipitate; 3) adding a reducing agent, which can break a disulfide bond,to the resuspension liquid and heating the resuspension liquid for 10-60 min at 37-80 DEG C; 4) adding a cleaning buffer solution to the solution in the step 3) and performing high-speed centrifugation for a certain time, and rejecting the supernatant and maintaining a precipitate; 5) re-dissolving the precipitate in the step 4) with a digestive buffer solution, and performing in-solution enzymolysis to proteins or performing protein separation through one-dimensional electrophoresis (SDS-PAGE). The method simplifies preparation process of urinary protein and improves accuracy and repeatability of detection. The method can be used for high-throughput quantitative deep detection of the urinary proteome.

The invention discloses a method for high-flux assay of phytase activity in a transgenic phytase crop seed. The method comprises the following steps: (1) building a phosphorus standard curve based on a phosphorus content as a horizontal coordinate and a light absorption value as a vertical coordinate; (2) leaching out phytase in a sample to be assayed so as to obtain leach liquor of phytase; (3) carrying out enzyme activity assay reaction of phytase on the leach liquor of phytase in a microplate with 96 holes; and (4) assaying the light absorption value of a reaction solution, calculating the phosphorus concentrations of various samples to be assayed according to a linear regression equation corresponding to the phosphorus standard curve, calculating the phytase activities of the samples to be assayed according to an enzyme activity calculation formula. Accuracy and sensitivity testes show that the assayed result obtained by using the method has good accuracy, small error, high stability and good repeatability, and coefficient of variation of the assayed result obtained by using the method is far lower than that obtained by the existing microplate method. The assay method disclosed by has the advantages of high flux, high assay efficiency, low cost, high accuracy and the like, and is easy to operate.
