The mass spectrometry method comprises the following steps: (1) spraying a solvent for desorbing target molecules into a purified conical through hole in a spray form of a spray, enabling the spray to run through the conical through hole, desorbing the target molecules to obtain a secondary spray comprising the target molecules, enabling the secondary spray to enter a sample feeding port of a mass spectrometer so as to finish sample feeding of one conical through hole, and similarly finishing sample feeding of a next conical through hole so as to finish mass spectrometry on each conical through hole by the mass spectrometer; (2) performing sample collection and data analysis by the mass spectrometer so as to acquire a mass spectrum image of component ions. The mass spectrometry method adopts the single-pixel solid-phase extraction technology innovatively; through intra-pixel point pretreatment, an inhibiting phenomenon of a complex matrix on the ions of the target molecules is effectively avoided, so that influence of complex matrix interference on the accuracy of the mass spectrum image is practically avoided and the sensitivity of low-abundance component imaging is improved.
