The invention relates to the field of textiles, in particular to a solvent method based high-wet-modulus bamboo, wood and cotton mixed fiber and a preparation method thereof. The preparation method includes steps: activating, adding bamboo, wood and cotton mixed pulp into deionized water, adjusting a pH value, adding cellulase for activating, and adding alkali for adjusting the pH value; pressing;pre-dissolving, and adding water solution containing N-methylmorpholine-N-oxide in mass percent of 50-88%; dissolving, heating, vacuumizing, dewatering, dissolving, homogenizing and de-aerating; filtering; spinning, and adopting a dry-wet method for spinning forming; washing; bleaching; oiling; drying. The preparation method is simple in operation, free of industrial pollution, low in energy consumption, high in safety and suitable for large-scale industrial continuous production of solvent method based bamboo, wood and cotton mixed fibers. The bamboo, wood and cotton mixed fiber prepared according to the method keeps natural physical and chemical performances of bamboo, wood and cotton fibers and is free of chemical residues and high in wet modulus.
