chip is used for constructing the flow micro-channels with the reducing structures in different appearance shapes, accurate and stable flow speed control is realized by virtue of a pneumatic
perfusion system, and controllable
shear stress is applied to the
neutrophile granulocytes through the reducing structures of the micro-fluidic
chip, so that the
drug-free pre-activation of the
neutrophile granulocytes is realized, and the
drug-free pre-activation of the
neutrophile granulocytes is realized. The antibacterial functions of
chemotaxis,
phagocytosis, NETs formation, ROS release and the like are directionally enhanced. The activated neutrophil can be used for treating local or systemic infection through local smearing or venous transfusion, and can be used independently or combined with
antibiotics. According to the method, the bacterial
clearance rate in an infection model can be remarkably increased, organ injury is relieved, and immune overreaction is regulated. The invention provides a novel
cellular immunotherapy for treatment of
drug-resistant bacterium infection and
immunodeficiency patients, and has standardized and repeatable clinical application potential.
