The invention discloses a preparation method of a chemically assisted cross-linked
chitosan temperature-sensitive gel submucosal injection for
endoscopic submucosal dissection, and belongs to the technical field of
regenerative medicine. According to the specific method,
chitosan, beta-
sodium glycerophosphate,
sodium dihydrogen
phosphate,
acetic acid, a chemical auxiliary cross-linking agent and a medical coloring indicator are utilized to construct the novel chemical auxiliary cross-linked
chitosan temperature-sensitive gel submucosal injection. The injection is very low in
viscosity, does not need a special
injection device, and can be injected by using a conventional
endoscope injection needle which is clinically used at present. Meanwhile, after the injection is injected under the
mucous membrane layer, high-strength hydrogel can be rapidly formed under the action of body temperature, the upheaval state of the
mucous membrane layer can be stably supported, ESD operation is facilitated, and the risk of perforation and bleeding is reduced. Particularly, a synergistic dynamic
proton ionization response system is established in the injection by
sodium dihydrogen
phosphate and
acetic acid, so that the
deprotonation speed of chitosan is remarkably slowed down, and the chemical auxiliary crosslinking speed is slow, so that the chemical auxiliary crosslinking does not influence injection,
mucous membrane lifting and electric resection of the injection in the ESD operation, and the crosslinking degree is gradually improved after the ESD operation. The chemical auxiliary crosslinking can significantly improve the adhesion stability of the gel layer on the postoperative
wound surface, form a stable
wound surface sealing protection gel layer, and effectively
delay the degradation of the gel layer
in vivo, especially in the gastric juice environment, so that the
adverse effect of the external environment on the postoperative
wound surface healing can be effectively reduced, and the postoperative wound surface healing effect is improved. And the degradation of the gel layer is more matched with the
tissue repair, so that the
wound healing is promoted. Therefore, the chemically assisted cross-linked chitosan temperature-sensitive gel submucosal injection has very important clinical practical application value.
