Product quality enhancement in mammalian cell culture processes for protein production

The present invention describes methods and processes for the production of proteins, particularly glycoproteins, by animal cell or mammalian cell culture, illustratively, but not limited to, fed-batch cell cultures. The methods comprise feeding the cells with D-galactose, preferably with feed medium containing D-galactose, preferably daily, to sustain a sialylation effective level of D-galactose in the culture for its duration, thus increasing sialylation of the produced proteins. The methods can also comprise at least two temperature shifts performed during the culturing period, in which the temperature is lower at the end of the culturing period than at the time of initial cell culture. The cell culture processes of the invention involving two or more temperature shifts sustain a high cell viability, and can allow for an extended protein production phase. The methods can also comprise the delayed addition of polyanionic compound at a time after innoculation. Supplementation of the cultures with D-galactose, preferably in a feed medium, to sustain galactose at sialylation effective levels in the cultures until the end of a culture run reverses a decline in sialylation that accompanies culture scale up, and is advantageous for large scale culturing processes.

Sample data processing method and device and computer readable storage medium

The invention discloses a sample data processing method and device and a computer readable storage medium. The method comprises the steps that a classification model is acquired; automatic annotationof model retraining sample data is performed through the classification model; the model retraining sample data and an automatic annotation result are displayed, the selection of correcting the displayed automatic annotation result is received, and an annotation expansion result of the model retraining sample data is obtained; and the model retraining sample data and the annotation expansion result are fed back to the model for training till the classification model no longer obtains classification performance improvement and iterative optimization of the annotation result corresponding to themodel retraining sample data is performed, and the error correction and leakage finding process of the corresponding annotation result is completed. In this way, annotation accuracy is improved, thesample data is continuously expanded, the corresponding automatic annotation result is obtained after automatic annotation through the classification model, the more precise annotation expansion result is obtained through correction, and the sample data in a large scale and large-quantity wide-dimensional annotation performed on the sample data are obtained.

Simulation data accessing method based on power grid CIM (Common Information Model) interface

The invention provides a power grid CIM (Common Information Model)-based method for accessing real-time data state estimated result of an EMS (Energy Management System) into an interface of a full-digital real-time simulation system. The method comprises the steps of: outputting a power grid model of the EMS and a telemetry information-equipment relation table into a format data file according to a CIM derivation standard, reading the CIM file in by a full-digital real-time simulation system of a power system through a data interface to form a computation database, and directly transmitting the power system simulation data based on the database back to the EMS for detection. The power grid CIM-based method has the advantages of multi-system joint debugging, high data precision and fast transmission speed, provides reliable data basis and unified network model for online safety analysis and prewarning control of a large power grid for the first time, and breaks through the bottleneck that calculation results cannot be compared because the offline calculation and a calculation module are inconsistent; and tests prove that a measurement model which is used for the full-digital real-time simulation system of the power system and constructed on the basis of the CIM derivation standard of the EMS is correct and consistent.

Hierarchical component based object recognition

The present invention provides a method for the recognition of objects in an image, where the objects may consist of an arbitrary number of parts that are allowed to move with respect to each other. In the offline phase the invention automatically learns the relative movements of the single object parts from a sequence of example images and builds a hierarchical model that incorporates a description of the single object parts, the relations between the parts, and an efficient search strategy. This is done by analyzing the pose variations (e.g., variations in position, orientation, and scale) of the single object parts in the example images. The poses can be obtained by an arbitrary similarity measure for object recognition, e.g., normalized cross correlation, Hausdorff distance, generalized Hough transform, the modification of the generalized Hough transform, or the similarity measure. In the online phase the invention uses the hierarchical model to efficiently find the entire object in the search image. During the online phase only valid instances of the object are found, i.e., the object parts are not searched for in the entire image but only in a restricted portion of parameter space that is defined by the relations between the object parts within the hierarchical model, what facilitates an efficient search and makes a subsequent validation step unnecessary.
