Gastro-retentive drug delivery system

The invention relates to floating drug delivery systems (FDDS) that provide solutions to the particular problems often encountered with floating drug delivery systems described in the art. On such generally recognized problem is the vulnerability of the systems, especially damage to the gas-filled compartment making it accessible to water so as to impair its buoyancy, ultimately resulting in insufficient gastric residence time. The invention, in an aspect, provides a self-repairing FDDS that maintains its floating capacity after damaging. The floating drug delivery systems of the invention, furthermore, allow for incorporation of high loads of active ingredients. The floating drug delivery systems can be designed in such a way that release of active ingredient from the system occurs entirely independent from the pH of the fluid surrounding the system. Furthermore, the procedure of manufacturing the floating drug delivery system of the invention is simple and straightforward, and therefore economically attractive.

Modified release drug powder composition comprising gastro-retentive raft forming systems having trigger pulse drug release

An orally administrable drug powder composition which forms a gastro-retentive RAFT having at least two trigger pulses is provided. The composition contains, at a minimum, (a) at least one drug in an immediate release pulse release form; (b) at least one drug in a delayed trigger release form; (c) at least one non-toxic gas generating agent and (d) a RAFT system, wherein following oral ingestion, the composition provides a self-assembling gastro-retentive RAFT having entrapped therein, the at least one drug of (a) and (b) and the gas generated in situ by the non-toxic gas generating agent, thereby providing a floating gastro-retentive RAFT having a dual pulse system wherein at least the second pulse is a trigger pulse and which retains the at least one drug in the stomach for at least about 3 hours, provided that the composition does not include a gamma hydroxybutyrate and its salts, hydrates, tautomers, or solvates, or complexes thereof.

Water-soluble drug gastroresistant coating granules, and preparation method and application thereof

The present application relates to the technical field of veterinary medicine preparation, and particularly relates to a water-soluble medicine gastro-resistant and enteric-coated granule, a preparation method and application thereof, the coated granule comprising: an inner core of at least one water-soluble medicine active ingredient, a coating layer coated outside the inner core; and the coating layer is formed by a composite coating material of hydrogenated palm oil and lipophilic hydrophobic nano active light calcium carbonate. The present application has the advantages that the nano calcium carbonate is used to form pores in the true stomach by acid reaction and to produce gas by saponification with fatty acids in the intestinal tract, so that the water-soluble medicine is efficiently protected in the rumen and rapidly and completely released in the intestinal tract. In vitro tests show that the effective release rate can reach more than 79%, which is significantly better than the traditional coating, and has a broad clinical application prospect.

Gastro-retentive sustained release formulations of amoxicillin

The present application relates to a kind of amoxicillin gastric retention sustained-release preparation, which contains amoxicillin, sustained-release material, swelling agent and other pharmaceutical adjuvant, preferably also contains stabilizer or ion regulator. The combination of the above-mentioned adjuvant can be retained in the stomach for up to 24 hours, the complete release time is 10-24h, which can significantly prolong the retention time of amoxicillin in the stomach, has the advantages of fast floating, high effective concentration of drug in the stomach, long stable concentration time, and is beneficial to reduce the frequency of drug administration of patients;Further preferably, it is made into a double-layer tablet containing immediate-release layer, which can quickly reach effective concentration and also can be released slowly and continuously, maintaining effective concentration for a long time; When combined with proton pump inhibitors such as vonolabian, etc., combination kit or compound, only 1-2 times per day is needed, which can further improve the therapeutic effect and patient compliance.

This invention discloses an extracellular vesicle derived from Gastrodia elata, its preparation method, and its use in preparing drugs for treating glaucoma, belonging to the field of biomedical technology. This invention claims protection for an extracellular vesicle prepared from dried Gastrodia elata. The extracellular vesicles provided by this invention have no significant cytotoxicity and good biocompatibility. Studies have shown that these extracellular vesicles can significantly inhibit apoptosis of R28 cells and loss of Brn3a⁺ cells in the mouse retina in a glutamate excitotoxicity model. In a glaucoma model, these extracellular vesicles can effectively protect the survival of retinal ganglion cells (RGCs) and retinal function, and can significantly reduce retinal neuroinflammatory responses in the glaucoma model. Therefore, the extracellular vesicles provided by this invention have a clear anti-glaucoma-related retinal damage effect and possess great potential for development into drugs for treating glaucoma.

An intelligent health patch system based on stomach-brain axis communication and a regulation method thereof

This invention relates to the field of intelligent medical device technology, specifically to an intelligent health patch system and its control method based on gastro-brain axis communication. The system includes an intelligent patch integrating a multimodal physiological sensing module for collecting gastric electrical signals and heart rate variability signals, and a biomimetic control execution module for performing transcutaneous electrical stimulation and tactile feedback; a mobile computing terminal configured with a gastro-brain axis state assessment model for evaluating the user's hunger drive level based on the physiological signals, and a personalized control decision engine for generating control instructions. The mobile computing terminal can, based on the output of the assessment model, achieve non-invasive closed-loop control of the gastro-brain axis: for the first time, closed-loop management of "ascending signal sensing" and "descending neural control" of the gastro-brain axis is realized on a consumer-grade wearable patch, making the intervention more physiologically targeted and scientific.
