Fenugreek seed bio-active compositions and methods for extracting same

A composition of bio-active compounds and methods for facilitating and supporting the metabolism and transport of glucose and carbohydrates into muscle cells. Preferably, the composition of bio-active compounds include 4-hydroxyisoleucine and one or more amino acids selected from the group consisting of arginine, aspartate, threonine, serine, glutamate, proline, glycine, alanine, cysteine, valine, methionine, isoleucine, leucine, tryptophan, phenylalanine, ornithine, lysine, histidine, gamma-amino butyrate, and trimethylhistidine. In one presently preferred embodiment of the present invention, the bio-active compounds are extracted from fenugreek seeds. A method for extracting a composition of bio-active compounds from fenugreek seeds is also disclosed, wherein the method comprises the steps of: (1) providing a plurality of fenugreek seeds; (2) preparing the fenugreek seeds; and (3) extracting a composition of bio-active compounds from the fenugreek seeds, wherein the bio-active compounds comprise 4-hydroxyisoleucine and one or more amino acids selected from the group consisting of arginine, aspartate, threonine, serine, glutamate, proline, glycine, alanine, cysteine, valine, methionine, isoleucine, leucine, tryptophan, phenylalanine, ornithine, lysine, histidine, gamma-amino butyrate, and trimethylhistidine. The composition of bio-active compounds and methods for extraction of same preferably include between about 10% and 70% of 4-hydroxyisoleucine and between about 20% and 40% of the amino acids.

α2-Heremans Schmid Glycoprotein (AHSG) inhibits insulin-induced autophosphorylation of the insulin receptor (IR) and IR-tyroskine kinase (TK) activity; genetic ablation of the Ahsg gene enhances insulin signal transduction and increase whole-body insulin sensitivity. Therefor, AHSG and its gene(s) are useful targets for agents that inhibit the development or progression of Type II diabetes or any disease or disorder associated with increased insulin resistance. Provided herein is a method for inhibiting the biological activity of AHSG protein in a cell using compounds that inhibit phosphorylation of AHSG. Also disclosed is a method of augmenting the phosphorylation or IR-TK activity in a liver or muscle cell by providing a compound that lowers the amount of active AHSG or inhibits the biological activity of AHSG. Such effects may be achieved by delivering an antisense nucleic acid construct that hybridizes with AHSG encoding DNA. This invention includes a method (a) treating a subject that is susceptible to, or suffers from, obesity and insulin resistance or (b) increasing insulin sensitivity, and thereby preventing or treating insulin resistance in the subject. The method comprises lowering the amount of active AHSG or inhibiting the biological activity of AHSG in the subject, preferably in liver or muscle, by using AHSG antisense constructs or an anti-AHSG antibody. In a subject eating a high fat diet, the effect on body weight gain and/or insulin resistance is diminished, and total body fat content is lowered, by lowering the amount of active AHSG or inhibiting the action of the AHSG in the subject using the agents noted above.

Compositions and methods for weight management

Compositions and methods for promoting weight management are taught by the present invention. Preferably, the methods include the step of administering a composition of bio-active compounds further including 4-hydroxyisoleucine and one or more amino acids selected from the group consisting of arginine, aspartate, threonine, serine, glutamate, proline, glycine, alanine, cysteine, valine, methionine, isoleucine, leucine, tryptophan, phenylalanine, ornithine, lysine, histidine, and gamma-aminobutyrate. In one presently preferred embodiment of the present invention, the bio-active compounds are extracted from fenugreek seeds. The methods of the present invention preferably include a composition of bio-active compounds with between about 10% and 70% of 4-hydroxyisoleucine and between about 20% and 40% of the amino acids. In presently preferred embodiments of the present invention, methods may work independently of or synergistically with insulin to stimulate the activity of glucose transport receptors located in muscle. More particularly, methods of the present invention may stimulate the activity of glucose transport receptor type 4. In another presently preferred embodiment of the present invention, a method for weight management includes the steps of: (1) performing a weight management analysis on an individual experiencing nutritional and metabolic deficiencies which impact proper metabolism of glucose and carbohydrates; (2) extracting a composition of bio-active compounds from fenugreek; and (3) administering the composition of bio-active compounds from fenugreek in an amount effective to enhance muscle glucose transportation within the body of the individual.
