The present invention provides a method of cryopreserving sperm that have been selected for a specific characteristic. In a preferred embodiment, the method is employed to freeze sex-selected sperm. Although the cryopreservation method of the invention can be used to freeze sperm selected by any number of selection methods, selection using flow cytometry is preferred. The present invention also provides a frozen sperm sample that has been selected for a particular characteristic, such as sex-type. In preferred embodiments, the frozen sperm sample includes mammalian sperm, such as, for example, human, bovine, equine, porcine, ovine, elk, or bison sperm. The frozen selected sperm sample can be used in a variety of applications. In particular, the sample can be thawed and used for fertilization. Accordingly, the invention also includes a method of using the frozen selected sperm sample for artificial insemination or in vitro fertilization.

The present invention provides an aptamer or pool of aptamers that bind(s) to a target molecule on the surface of a mammalian sperm cell and a method for producing the aptamers. The method comprises contacting a collection of different nucleic acid molecules with the target molecule under conditions favorable for binding between at least one of the nucleic acid molecules and the target molecule, to form at least one complex comprising the nucleic acid molecule bound to the target molecule, wherein each of the nucleic acid molecules comprises at least one segment of randomized nucleotide sequences. The complexes are then separated from the unbound nucleic acid molecules and unbound target molecules, and the bound nucleic acid molecule is recovered from the separated complex. The aptamers are used to separate sperm cells based upon sperm cell qualities, such as whether the cells contain X chromosomes or Y chromosomes.

A method for adjusting the maturation state of mammalian sperm for use in assisted reproductive technologies (ART) is disclosed. A mammalian ejaculate is provided and incubated under controlled conditions. Aliquots of the ejaculate are assayed during incubation period at intervals to determine maturation state and changes in the maturation state by observing the percent positive cells in the aliquot. The assays are repeated with successive aliquots at intervals during incubation to observe real time changes in the maturation state. The ejaculate remaining is processed for the desired ART after the percentage of positive cells in the latest aliquot being assayed begins to decline.
