Single-primer nucleic acid amplification methods

The present invention is directed to novel methods of synthesizing multiple copies of a target nucleic acid sequence which are autocatalytic (i.e., able to cycle automatically without the need to modify reaction conditions such as temperature, pH, or ionic strength and using the product of one cycle in the next one). In particular, the present invention discloses a method of nucleic acid amplification which is robust and efficient, while reducing the appearance of side-products. The method uses only one primer, the “priming oligonucleotide,” a promoter oligonucleotide modified to prevent polymerase extension from its 3′-terminus and, optionally, a means for terminating a primer extension reaction, to amplify RNA or DNA molecules in vitro, while reducing or substantially eliminating the formation of side-products. The method of the present invention minimizes or substantially eliminates the emergence of side-products, thus providing a high level of specificity. Furthermore, the appearance of side-products can complicate the analysis of the amplification reaction by various molecular detection techniques. The present invention minimizes or substantially eliminates this problem, thus providing an enhanced level of sensitivity.

Multi-focal treatment of skin with acoustic energy

Methods and apparatus are disclosed for applying acoustic energy to the skin. Acoustic waveguides with elements of varying thickness or shape are disclosed which deliver energy to more than one depth below a surface of the skin substantially simultaneously. The invention is especially useful with devices that focus ultrasound energy by condensing a propagating wavefront. The invention compensates for the mismatch in acoustic properties of the device's waveguide and the biological tissue that typically cause portions of the collapsing wavefront to lag behind other portions and, thereby, limit the focusing capabilities of acoustic treatment devices.

A stable topical alcohol-free aerosol foam containing one or more keratolytic agents is provided. The foam-forming formulation is an emulsion which contains an HFA propellant and one or more keratolytic agents. The emulsion has an oil phase and an aqueous, i.e. water-containing, phase. The active agent(s) may be present in either phase of the emulsion or dispersed in the emulsion. The oil phase may consist at least in part of the HFA propellant. Either or both of the oil phase and the aqueous phase may contain one or more surfactants, emulsifiers, emulsion stabilizers, buffers, and / or other excipients. The foam is stable on the skin, for example, for at least 5 minutes at body temperature, preferably at least 20 minutes at body temperature, and disappears into the skin upon rubbing or after prolonged standing. In one embodiment, the formulation contains an HFA propellant which does not contain additional co-solvents or co-propellants. The formulations demonstrate reduced intensity of the odor and / or color associated with the keratolytic agent(s) as compared to conventional formulations containing keratolytic agents.

Digital image system and method for implementing an adaptive demosaicing method

An adaptive demosaicing method interpolates images based on color edge detection and neighborhood voting. The adaptive demosaicing algorithm uses a voting scheme to determine the direction of interpolation at each missing luminance pixel location. Each color plane votes either horizontal or vertical based on a comparison between the horizontal and vertical components of the degree of change (i.e., gradient, Laplacian or other measure of the degree of change) in that color plane. Votes are counted from the neighborhood pixels as well as from measurements taken at the pixel location itself. Once the luminance plane is fully interpolated, the chrominance planes are filled in by simple bilinear or median interpolation of difference chrominance values. Enhancements to the adaptive demosaicing algorithm permit adaptive smoothing and sharpening.

A distal region of the probe is engaged with the target region. Measuring and recording current temperature of a proximal region of the probe and time of the measurement is used to determine slope of a temperature curve defined by two points. The first point is defined by the current temperature and time of measurement and a second point is defined by a previous measurement of proximal region temperature and time of measurement. When the slope is less than a slope threshold value a treatment flag is activated, treatment start time is recorded and the proximal region is heated with the heater element. Heating is discontinued and the treatment flag is deactivated after elapsed treatment time exceeds a duration threshold value.

A method and system for skin tightening comprises a hollow cannula that contains an optical fiber connected to a laser source. The cannula is inserted subcutaneously into a patient so that the end of the fiber is located within the tissue underlying the dermis. The source emits an output pulse that is conveyed by the fiber to the dermis, where the pulse causes collagen destruction and shrinkage within the treatment area. Radiation from the skin surface is detected to prevent non-reversible damage to the dermis, such as skin necrosis and excessive collagen melting. This method of sub-cutaneous laser treatment can also be used to treat striae, or stretch marks.
