This invention provides a dual-segment, dual-color pH comparison catheter for clinical identification of premature rupture of membranes, belonging to the field of medical catheter technology. It includes a main catheter tube with a urine segment pH membrane and an amniotic fluid segment pH membrane sequentially arranged along the axial direction on the anterior outer wall of the main tube. The urine segment pH membrane has a color development range of pH 5.5-6.5, and its color change is a continuous gradient from yellow to green. This invention achieves zero-sampling detection under sterile conditions, highly integrating pH identification function into routine catheterization procedures. The pH value of urine and any potentially mixed amniotic fluid is simultaneously detected upon catheter insertion into the bladder, without the need for vaginal speculum, sampling swabs, or any form of secondary sterile opening. This invention discloses the application of MIF gene silencing in the preparation of drugs for treating premature rupture of membranes (PPROM), belonging to the fields of biomedicine and molecular biology. By collecting fetal membrane tissues from patients with PPROM and normal controls, this invention found that the expression level of MIF protein and the concentration of PGE2 were significantly increased in the PPROM group. Further investigation using siRNA technology to silence the MIF gene in human amniotic epithelial cells revealed that MIF silencing significantly reduced PGE2 levels. This invention is the first to demonstrate that MIF gene silencing can effectively treat PPROM by reducing PGE2 (inhibiting uterine contractions). This invention also provides a pharmaceutical composition containing MIF siRNA. This invention provides a novel molecular target and therapeutic strategy for the treatment of PPROM, with broad clinical application prospects.
