Under the condition of pH triggering, the fluorescent probe LD-Nu-APP can dye the lipid droplets and the nucleolus and emit green fluorescence and red fluorescence respectively. The advantages of two-parameter detection (lipid content and nucleolus size) of the fluorescent probe LD-Nu-APP are utilized, the non-alcoholic fatty liver tissue and the normal tissue can be successfully distinguished, and the method is more accurate than Nile red which only depends on lipid change. Besides, based on the Bayesian theorem, the fluorescent probe LD-Nu-APP is used for screening and evaluating the two drugs for treating the non-alcoholic fatty liver disease, and it is found that pioglitazone is more effective than metformin, and the application range is wider. In conclusion, it is believed that the fluorescent probe LD-Nu-APP has unique lipid content-nucleolus size two-parameter detection characteristics and has huge value in non-alcoholic fatty liver disease diagnosis and drug screening.
