The invention discloses a silk
fibroin-based bionic artificial tympanic membrane with a gradient structure and a preparation method thereof, and the preparation method comprises the following steps: wrapping and dissolving an
antibacterial drug in an
organic solvent, then dropwise adding into a silk
fibroin solution,
casting to form a membrane, and then inducing by
methanol to form a beta-
folded structure to obtain an outer compact layer; carrying out electrostatic
spinning on the silk
fibroin solution to obtain a directionally arranged silk fibroin
fiber felt, and soaking and cross-linking in an
ethanol solution to obtain a middle
fiber layer; mixing the silk fibroin solution with a pore-
foaming agent, and pouring the mixture on the middle
fiber layer; covering the middle
fiber layer with the outer compact layer, pre-freezing and freeze-
drying, immersing into deionized water to dissolve out the pore-
foaming agent so as to form an inner
porous layer, putting into a PBS
buffer solution containing growth factors for oscillation treatment, and
drying, thereby obtaining the silk fibroin-based bionic artificial tympanic membrane with the gradient structure. The artificial tympanic membrane with the three-layer bionic structure has antibacterial and regeneration promoting functions, has excellent mechanical properties and meets the requirements of bionic
tympanic membranes.
