Chronic Rejection Inhibitor

ActiveUS20100061986A1Suppressing chronic rejection reactionAvoid spreadingAntibody ingredientsImmunoglobulinsHeart transplantationStenotic lesion
The present inventors assessed the effect of anti-IL-6 receptor antibodies in suppressing chronic rejection reaction. They assessed the effect of anti-mouse IL-6 receptor antibody (MR16-1) administration in suppressing the chronic rejection reaction using a mouse model for post-heart-transplantation chronic rejection. The result of histopathological analysis of transplanted hearts extirpated 60 days after transplantation revealed that fibrosis of myocardium and vascular stenotic lesions, which are pathological conditions characteristic of the chronic rejection reaction, were significantly suppressed in the MR16-1-treated group as compared to the control group. Thus, MR16-1 administration was demonstrated to have the effect of suppressing chronic rejection reaction. Specifically, the present inventors discovered for the first time that the rejection reaction in the chronic phase after organ transplantation was suppressed by administering an anti-IL-6 receptor antibody.

Accelerated evaluation of treatments to prevent clinical onset of alzheimer's disease

Systems and methods for evaluating prospective treatments for progressive brain disorders like Alzheimer's disease and the progressive effects of aging involve the use of an imaging device communicatively coupled to a computing device. The imaging device may takes a plurality of brain imaging measurements from each of a plurality of human subjects who are divided into a treated group and an untreated group based on various subject data including whether the subjects carry certain alleles of genes known to increase the risk of developing the brain disorder at issue. The computing device receives the brain imaging measurements from the imaging device and applies an advanced processing method that permits enhanced computational efficiency when calculating rates of change for the two groups and determining whether any calculated difference between the rates is statistically significant.

Biological agents active in central nervous system

Cell-permeant fusion peptides Tat-PDZ can dose-dependently reduce the threshold for anesthesia. PDZ domain-mediated protein interactions at synapses in the central nervous system play an important role in the molecular mechanisms of anesthesia. Moreover, Tat-PDZ cell-permeant fusion peptides are delivered intracellularly into neurons in the central nervous system subsequent to intraperitoneally injection. By in vitro and in vivo binding assays, we found that the Tat-PDZ dose-dependently inhibited the interactions between NMDARs and PSD-95. Furthermore, behavior testing showed that animals given Tat-PDZ exhibited significantly reduced established inflammatory pain behaviors compared to vehicle-treated group. Our results indicate that by disrupting NMDAR / PSD-95 protein interactions, the Tat-PDZ cell-permeable fusion peptides provide a new approach for inflammatory pain therapy.

Intranasal vasopressin treatment of social deficits in autism

Provided herein are methods for treating, reducing and / or ameliorating symptoms of autism spectrum disorder (ASD) in a subject, where at least one measure of social impairment, anxiety, or repetitive behaviors is treated and / or ameliorated. Aspects include intranasally administering arginine vasopressin (AVP), or an analog of vasopressin, to a subject (e.g., a child between 6 and 12 years of age and having endogenous pre-treatment levels between about 0.045 and about 4.028 pg / mL, where the mean was 1.324 pg / mL). In children having high pre-treatment blood levels of AVP (e.g., about 25% or more above the mean pre-treatment AVP blood levels of the treatment group; or having pre-treatment blood vasopressin levels greater than 1.324 pg / mL), the treatment was enhanced. Because no difference in pre-treatment blood AVP levels was observed between control and ASD subjects, it was surprising that AVP treatment was more effective in subjects having high pre-treatment AVP levels.
