The invention discloses an apolipoprotein A1 fusion protein for preventing and treating atherosclerosis as well as a preparation method and application of the apolipoprotein A1 fusion protein, and belongs to the technical field of biological medicines. The method specifically comprises the following steps: cloning an apolipoprotein A1 gene to a pET28a expression vector, and transforming escherichia coli for expression; the expression product is purified, purification conditions are optimized, and the physicochemical properties of the expression product are represented by related means. In addition, the invention also relates to application of the fusion protein in the cellular level, including cytotoxicity experiment, and evaluation of protein uptake capability and cholesterol reverse transport capability of cells. Experimental results show that the prepared apolipoprotein A1 fusion protein can specifically target the liver, the atherosclerotic plaque area is remarkably reduced, and the apolipoprotein A1 fusion protein has important application potential in prevention and treatment of atherosclerosis.
