DNA amplification and subtraction techniques

A method of isolating genomic or RNA-derived duplex fragments which are unique to one of two fragment mixtures. The fragments in positive-source and negative-source mixtures are separately equipped with end linkers, and each mixture is amplified by successive primed-strand replications, using a single primer which is homologous to the associated linker. The second-source linker is biotinylated, and the fragments in this mixture are hybridized in molar excess with the fragments in the positive-source mixture. DNA species which are not hybridized with the biotinylated species, i.e., species that are unique to the positive-source mixture, are isolated after removal of hybridized species by affinity chromatography. Also disclosed is a method of amplifying a mixture of DNA fragments by repeated linker / primer replication.

Said plants are functionally male-sterile and are useful for the production of hybrid seed by the crossing of said male-sterile plants with pollen from male-fertile plants. Hybrid seed production is simplified and improved by this invention and can be extended to plant crop species for which commercially acceptable hybrid seed production methods are not currently available.

Through observation of chloroplast by using a transmission electron microscope, the fact that the OscpSRP54 gene has the function of modulating chloroplast growth and then further controlling the leaf color is proved. The OscpSRP54 gene can be used to modulate paddy rice chloroplast growth so as to improve the photosynthesis efficiency and increase the paddy rice output. The leaf color control gene provided by the invention can also be used as a tracing marker in a genetic transferred plant progeny or an indicating marker for distinguishing pure species from hybrid species in a hybrid seed production process, and can be applied to the fields of good species breeding and hybrid breeding.
