The invention discloses a method for preparing egg white lysozyme and active protein by adopting coseparation. The method disclosed by the invention comprises the following steps of: collecting egg white at room temperature, regulating pH to be about 5.5, stirring to be uniform and then centrifuging; adding poly vinyl alcohol with the molecular weight of 200-20000 into supernate obtained by centrifuging, stirring to be uniform and then centrifuging; adding a saturated ammonium sulphate solution into the supernate obtained by centrifuging, mixing to be uniform and then centrifuging; respectively collecting supernate and subnatant which are obtained by centrifuging; carrying out ultrafiltration or dialysis treatment on the supernate to obtain high-purity egg white lysozyme; and carrying out ultrafiltration or dialysis treatment on the subnatant to obtain other high-purity active proteins which are ovalbumin and ovotransferrin mainly. The method disclosed by the invention is simple and practicable, cost is low, conditions are mild, and good quality of lysozyme can be maintained, so that the method disclosed by the invention is applicable to large-scale industrialization production; and other active proteins can also be obtained, so that production benefit is further increased.

The invention provides a method for establishing a tuberculosis risk predicting model based on an iron metabolism index. The method comprises the following steps of (1), collecting and processing a sample; (2), measuring the iron metabolism index from the sample, particularly the iron metabolism comprises data of three kinds of serum iron, namely serum iron, transferring and ferritin; (3), fittinga training set by means of a neural network method for establishing the model, recording an optimal model parameter; calculating a model classifying threshold according to an ROC curve by means of averifying set, and establishing the tuberculosis risk predicting model. According to the method of the invention, data of the iron metabolism index in the sample human serum is measured; modeling analysis is performed on the three indexes by means of a neural network model; and an algorithm and the iron metabolism indexes are used for establishing a method for predicting the tuberculosis risk. Themodel has high sensitivity and high specificity and can be used for risk evaluation of the tuberculosis.

By using DMEM (dulbecco's modified eagle medium) / F12 as a basal culture medium, micromolecule substance iron salt with definite component, zinc salt, sodium selenite, sodium acetate, glutamine and glucose are added according to the characteristics of the microencapsulated cell culture. The culture medium does not influence the stability of the microcapsules, does not have insulin or transferrin in the traditional serum-free culture medium, and lowers the downstream technique and cost; and the product expression level is approximate to the culture level of the expensive specific culture medium, but the price is only one-tenth of the expensive specific culture medium. Thus, the protein-free culture medium can be used for microencapsulated recombinant CHO cells culture and protein expression, and greatly lowers the culture cost.

NTBI appears in serum when there is excess iron in the body. The method comprises of employing a signal generating moiety capable of complexing with iron that is a peptide like molecule having an iron binding site and also an optical signal generating functional group. The molecule is of microbial origin. The measurement is based on the alteration of optical characteristics of the probe molecule upon attachment of iron to its binding site on the molecule. Hence it generates a signal proportionate to the amount of iron available for binding and provides a direct estimate of free or unbound iron in the sample. According to this instant invention a rapid estimation method of NTBI in body fluids can be undertaken in an inexpensive way without the need of specialized expertise.
