AXL inhibitors for the treatment of mastocytosis

Mastocytosis is a rare and heterogeneous disorder marked by the abnormal accumulation of mast cells (MCs) in various tissues. Tyrosine kinase inhibitors have shown limited efficacy, suggesting that additional molecular mechanisms are involved. The inventors showed the unexpected expression of AXL in neoplastic mast cells from patients with various forms of mastocytosis. This is particularly noteworthy as AXL expression had never been investigated in mast cells, whether neoplastic or normal. The study demonstrated the role of AXL, particularly in cooperation with KIT D816V to drive mast cell proliferation and resistance to therapies. In an embodiment, the AXL inhibitor bemcentinib (R428) is used in combination with the TKI midostaurin (PKC 412). The AXL-L197M identified in the ASM patient is pro-proliferative, and induces resistance to death in CD34-derived mast cells and in Ba-f3. Furthermore, the results also suggest that a combination of AXL inhibitors and BCL-2 inhibitors may be appropriate for the treatment of mastocytosis. In conclusion, AXL inhibitors can be thus suitable for the treatment of mastocytosis.

An antiallergic pharmaceutical composition, a preparation method thereof, and use thereof

This invention discloses a fusion peptide with synergistic anti-allergic effects, an anti-LTB4 monoclonal antibody, and an anti-allergic drug composition thereof. The fusion peptide is composed of peptide A, which inhibits histamine release, and peptide B, which blocks mast cell activation signals, covalently linked by a linker peptide, which can synergistically inhibit mast cell activation. The anti-LTB4 monoclonal antibody can specifically bind to LTB4, blocking its mediated inflammatory response. Combining the fusion peptide, the anti-LTB4 monoclonal antibody, and myricetin, which has antioxidant activity, forms a multi-target drug composition that can synergistically inhibit allergic reactions from multiple aspects, such as inhibiting mast cell degranulation, blocking the action of inflammatory mediators, and clearing oxidative stress products. In vitro cell experiments and OVA-induced allergic asthma mouse models have demonstrated that the anti-allergic effect of this composition is significantly better than that of a single component, and it has no obvious toxicity, providing a new candidate drug for the efficient treatment of allergic diseases.

This invention provides a mast cell-targeting magnetoelectric hydrogel acupuncture system for acupoint stimulation, its preparation method, and its application, belonging to the technical field of biomedical materials and acupuncture-assisted treatment devices. The invention first synthesizes Fe₃O₄@BaTiO₃–FcεRIA (FBTFA) nanoparticles to achieve mast cell targeting and synergistic stimulation of cells by "mechanical force-electrical energy" under magnetic field induction.
