Interstitial microwave antenna with miniaturized choke hyperthermia in medicine and surgery

The present invention relates to minimally invasive surgery techniques. It provides a method for manufacturing an antenna for percutaneous acute hyperthermia microwave applications of the monopole or dipole co-axial type provided with trap, commonly called choke, for blocking the propagation of the backwards reflecting wave towards the generator. The miniaturization of the device allows a use minimally invasive for interstitial hyperthermia in medicine and surgery, in particular for oncology. The method of manufacturing the antenna provides a metal needle (1) for the introduction of the antenna (2,3,4) in the target tissue. On the external conductor (4) of the antenna (2) a metal collar 6 is connected in a predetermined position; a plastics sheath (5) is applied in order to cover the external conductor (2) in the portion between the feed (7) and the collar 6; the inner wall of the metal needle 1 wherein the antenna is inserted is then used for containing and guiding the collar 6 and the sheath 5; in particular, the collar 6 being in electrical contact with the inner wall of the metal needle 1. An antenna is thus obtained with choke of variable length and with miniaturized diameter. A thermocouple can be introduced through the choke that protrudes the directly in the “feed” zone.

Humanized anti-CD22 antibodies and their use in treatment of oncology, transplantation and autoimmune disease

The present invention provides chimeric and humanized versions of anti-CD22 mouse monoclonal antibody, HB22.7. The anti-CD22 antibodies of the invention comprise four human or humanized framework regions of the immunoglobulin heavy chain variable region (“VH”) and four human or humanized framework regions of the immunoglobulin light chain variable region (“VK”). The invention further comprises heavy and/or light chain FW regions that contain one or more backmutations in which a human FW residue is exchanged for the corresponding residue present in the parental mouse heavy or light chain. Human or humanized VH framework regions of antibodies of the invention may comprise one or more of the following residues: a valine (V) at position 24 of framework region 1, a glycine (G) at position 49 of framework region 2, and an asparagine (N) at position 73 of framework region 3, numbered according to Kabat. The invention further relates to pharmaceutical compositions, immunotherapeutic compositions, and methods using therapeutic antibodies that bind to the human CD22 antigen and that preferably mediate human ADCC, CDC, and/or apoptosis for: the treatment of B cell diseases and disorders in human subjects, such as, but not limited to, B cell malignancies, for the treatment and prevention of autoimmune disease, and for the treatment and prevention of graft-versus-host disease (GVHD), humoral rejection, and post-transplantation lymphoproliferative disorder in human transplant recipients.

Preparation method for soluble truncated human tumor necrosis factor-related apoptosis inducing ligand (TRAIL) active protein

The invention provides a preparation method for a soluble truncated human tumor necrosis factor-related apoptosis inducing ligand (TRAIL) active protein, comprising the following steps of: (A) designing and synthetizing an oligomerization deoxyribonucleic acid (DNA) single-stranded segment according to the amino acid sequence of the human TRAIL protein issued by a Genebank by the preference of a bacterium genetic code; (B) synthetizing a complete double stranded DNA by three-time polymerase chain reaction (PCR); (C) carrying out amplification by utilizing a T-carrier, and inserting the amplified double stranded DNA segment into an expression carrier and screening a positive transformant; (D) expressing the truncated human TRAIL protein in escherichia coli at low temperature; (E) purifying the protein by using a three-step method; and (F) determining the truncated TRAIL protein. The invention realizes that the truncated human TRAIL protein is efficiently expressed in an escherichia coli cell and a purification preparation technique thereof and overcomes the problem that an infusible inclusion body is easy to form in the prior art. The method has the advantages of simple operation, short fermentation time and easy purification, and moreover, the protein is ensured not to have any modification, and the protein reaches electrophoretically pure. The invention can be directly used for the research work of biochemistry and molecular biology and the oncology and the preclinical stage of tumor treatment or the development of clinical drugs.
