The present application belongs to the technical field of animal feed preparation, and particularly relates to a straw-based single-cell protein feed as well as a preparation method and application thereof. In the present application, steam explosion is combined with alkali-oxygen pretreatment to break the straw lignin barrier and improve the polysaccharide degradation efficiency; a mutant of Trichoderma reesei is obtained by using atmospheric and room temperature plasma (ARTP) mutagenesis, and is used for solid-state fermentation in combination with bacteria and enzymes to efficiently convert fermentable sugar and greatly improve the crude protein content of the product. The process abandons traditional high-pollution chemical treatment, is water-saving and environmentally friendly, uses cheap and easily available raw materials, and is easy to scale up. The present application can increase the crude protein content of straw from 4% to 6% to 10% to 15%, and the upgrading effect is remarkable. The feed obtained by the present application can replace 20% to 30% of the crude protein raw materials in the feed, improve the growth performance and nutrient digestibility of animals, and realize the resource-based high-value utilization of straw.
