The method includes the following steps that (1) by means of moderate microclimate conditions under protected cultivation frame overhead planting bed, ground under the bed serves as a dendrobium temporary planting seedbed; (2) culture medium main raw materials are fermented; (3) culture media are prepared and put into a seedling hole plate; (4) the dendrobium tissue culture seedlings of the right age are selected for indoor seedling hardening; (5) the tissue culture seedlings are placed on the temporary planting seedbed after temporary planting on the seedling hole plate; (6) management after temporary planting is carried out. After the method is adopted, the temporary planting survival rate of the dendrobium tissue culture seedlings is high, the cultured hole plate seedlings are strong and uniform, seedling survival is easy after planting, growth is quick, the method is suitable for seedlings for dendrobium protected cultivation and bionic cultivation large-scale production, and the utilization rate of equipment can be improved.
