Cucurbita (Latin for gourd) is a genus of herbaceous vines in the gourd family, Cucurbitaceae, also known as cucurbits, native to the Andes and Mesoamerica. Five species are grown worldwide for their edible fruit, variously known as squash, pumpkin, or gourd depending on species, variety, and local parlance, and for their seeds. Other kinds of gourd, also called bottle-gourds, are native to Africa and belong to the genus Lagenaria, which is in the same family and subfamily as Cucurbita but in a different tribe. These other gourds are used as utensils or vessels, and their young fruits are eaten much like those of Cucurbita species.

The test strip can be used for detecting the CGMMV in a cucurbitaceous plant and is applicable to detection and diagnosis of a viral disease caused by the CGMMV. The method is simple and quick. The test strip is low in cost. A result can be obtained within 5 to 10 minutes and can be visually and directly observed by naked eyes. The test strip is applicable to quick and extensive screening and detection of diseases through basic agricultural departments at all levels, farmers and entry-exit inspection departments, facilitates timely and effective control and elimination of an epidemic situation, and has a wide application prospect.

The method is characterized by designing gynostemma pentaphylla SCAR marked specific primer pair WJS-T1 and WJS-T2 by using the result of medicinal plant cucurbitaceae gynostemma pentaphylla DNA characteristic segment SCAR marked clone sequencing, designing internal reference primer pair WHL-18S-C1 and WHL-18S-C2 according to a cucurbitaceae gynostemma pentaphylla 18S rRNA conserved region, forming a polymerase chain reaction (PCR) composite system 1, and amplifying the cucurbitaceae gynostemma pentaphylla DNA by using a touchdown PCR program to obtain two zones, namely a 359bp zone and a 293bp zone; anddesigning the internal reference primer pair WPT-18S-C1 and WPT-18S-C2 according to a vitaceae 18S rRNA conserved region, combining the internal reference primer pair WPT-18S-C1 and WPT-18S-C2 and the gynostemma pentaphylla SCAR marked specific primer pair WJS-T1 and WJS-T2 to form a PCR composite system 2, and amplifying a vitaceae cayratia japonica DNA by using the touchdown PCR program to obtain an 18S rRNA 177bp zone. Therefore, the DNA level of medicinal plant cucurbitaceae gynostemma pentaphylla is identified, and the distinction between the gynostemma pentaphylla and the vitaceae cayratia japonica which is easy to confound with the cucurbitaceae gynostemma pentaphylla in shape is made.
