The invention provides a method for measuring liquid phase ozone concentration by flow injection ozonation, which solves the problems of measurement result error caused by selection of traditional trapping agents and interference of metal ions in matrix water samples, and secondary pollution in the prior art. The method comprises the following steps: (1) conveying an ozone solution to be detected through a pump; (2) mixing the ozone solution with a tertiary butanol solution in a tertiary butanol pipeline; (3) mixing with a luminol solution in a luminol pipeline and a metal-chelator solution in a metal-chelator solution pipeline, detecting chemiluminescent signals generated by ozone molecules and luminol in the mixed solution by a photomultiplier; (4) collecting and amplifying the optical signals, converting into electric signals, sending the electric signals into a computer data processing system, calculating the liquid phase ozone concentration, displaying, and printing for output. The tertiary butanol is adopted to eliminate interference on luminol ozonation caused by traditional trapping agents; the metal-chelator is adopted for complexation with metal ions in the water, and the generated metal complex does not catalyze chemiluminescence of luminol.

In order to solve the above-mentioned problem, the present inventor has found that in a cell-free protein synthesizing system, in particular, a wheat embryo cell-free protein synthesizing system, when biotinylation is performed during or after protein's synthesis, the biotinylation of the protein can be attained in an remarkably lower concentration of the biotin than that in the conventional biotinylation operations, and has accomplished the present invention by using the protein having the biotin tag in each detection system.

Method for measuring water sulfide concentration through flow injection chemiluminescence

The invention provides a method for measuring a water sulfide concentration through flow injection chemiluminescence. The method is performed according to the following steps of: 1. inputting a measured water sample into a water sample reaction chamber in a reaction chamber; 2. delivering ozone into the water sample reaction chamber in the reaction chamber, performing a gas-liquid phase reaction for 30-50s, carrying out ozone oxidation to the sulfide in the water sample so as to generate sulfur dioxide; 3. transporting the completely reacted measured water sample solution, which is mixed with phosphoric acid in a phosphoric acid pipeline; 4. then mixing the mixed solution mixed with the phosphoric acid with a potassium permanganate solution, and letting a photomultiplier tube detect a chemiluminescent signal generated during oxidation of the sulfur dioxide by potassium permanganate in the solution; and 5. allowing the photomultiplier tube to collect and amplify an optical signal emitted by a solution flowing through, converting the signal into an electric signal, which is then sent into a microcomputer data processing system, then quantizing the signal, and calculating the concentration of the sulfide in the water. The invention has the advantages of high sensitivity, wide linear range, rapidity, good reproducibility, as well as high degree of automation, etc.

Mismatch repair detection

Mismatch Repair Detection (MRD), a novel method for DNA-variation detection, utilizes bacteria to detect mismatches by a change in expression of a marker gene. DNA fragments to be screened for variation are cloned into two MRD plasmids, and bacteria are transformed with heteroduplexes of these constructs. Resulting colonies express the marker gene in the absence of a mismatch, and lack expression in the presence of a mismatch. MRD is capable of detecting a single mismatch within 10 kb of DNA. In addition, MRD can analyze many fragments simultaneously, offering a powerful method for high-throughput genotyping and mutation detection.
