Tissue equivalents

A method of forming a tissue equivalent is described. A length of polyester tube is threaded over a mandrel (H) and attached at either end by clips at A and D. A block (F) is then screwed into place. The polyester is then pre-soaked by injecting through Tap (2) an acidified collagen solution for approximately one hour. After a suitable period of time the excess solution is aspirated off. Following this stage, a second, alkaline solution is injected into the apparatus which contains smooth muscle cells (SMC). Thus, neutralisation of the collagen impregnated within the fabric of the tube occurs leading to spontaneous fibrillogenesis within the interstices of the cloth, eliminating the risk of delamination. The apparatus is then incubated. The collagen contracts down onto the fabric tube and the cell-impregnated gel becomes incorporated into the presoaked collagen. The pre-impregnated collagen and the collagen provided in the aqueous mixture contract down as one into a coherent whole with the SMC. The tissue equivalent is then lined with endothelial cells and the apparatus again incubated and fed at increasing intraluminal pressures applied either statically or dynamically. This is believed to cause the vessel to become preconditioned to the pressure it will work under as an implant. This allows organization of the basement membrane which in turn promotes EC attachment and theoretically prevents smooth muscle cell hyperplasia.

Method for detecting tobacco-specific nitrosamines

The invention relates to a method for measuring tobacco-specific nitrosamines (TSNAs for short) by using a super efficient liquid chromatogram-tandem mass spectrum combined method, and belongs to a method for detecting TSNAs. The method comprises the following steps of: adding internal standard-containing aqueous solution of ammonium acetate into a filter sheet after trapping smoke and cigarette filters smoked completely under standard conditions or tobacco shreds, performing ultrasonic extraction, transferring 1 to 2mL of extract into a small solid-phase extraction column taking N-vinyl pyrrolidone-benzene sulfonic acid copolymer as filler, flushing the small column by using aqueous solution of methanoic acid and methanol, finally eluting the small column by using solution of aqueous ammonia and methanol solution, analyzing the eluant by using an ultra performance liquid chromatography-mass spectrometer/mass spectrometer (UPLC-MS/MS), and quantifying according to the area ratio of a component peak to an isotope internal standard peak. The detecting method can be used for detecting the TSNAs in cured and hybrid cigarette mainstream smoke, cigarette side-stream smoke, filter tip intercepting smoke and tobacco shreds, and has the advantages of simple pretreatment, accurate quantification, high instrument analysis speed, low detection limit, high sample treatment flux and the like.

The invention discloses a three-dimensional mesh model tetrahedralization method. The method comprises the following steps that (1), the initial position of a model is preprocessed; (2) a body centered cubic structure is established; (3) node symbols and points of tangency of boundries of a tetrahedron with the node symbols at the two ends opposite are calculated; (4) the points of tangency are moved; (5) the boundaries are tetrahedralized again. According to the three-dimensional mesh model tetrahedralization method, based on a body-centered cubic mesh tetrahedralization algorithm, preprocessing of principal component analysis of the three-dimensional model is added, a movement mode of the points of tangency of the boundaries of the model is changed, the points of tangency are moved to feature points of the model, and the positions of nodes of a final tetrahedral mesh are optimized through a density energy error function. After principal component analysis is adopted in the three-dimensional model, the quality of an initial tetrahedron element is improved; movement of the points of tangency to the feature points of the model is improved, so that local features of the tetrahedralized model are guaranteed; the quality of the final tetrahedral mesh is optimized through the density energy error function.
