The application discloses application of MANF in preparation of a
medicine for treating
intervertebral disc degeneration, and belongs to the technical field of medicines. The
medicine comprises a
midbrain astrocyte-derived neurotrophic factor or a
reagent for overexpressing MANF, the
reagent is a
gene for expressing MANF, and the
medicine further comprises a
lentivirus vector for carrying the
reagent for overexpressing MANF. Expression of MANF is evaluated by using human
nucleus pulposus tissue and a rat
intervertebral disc puncture model,
in vitro, a simulated degeneration condition of
acid treatment is adopted, and the MANF level in
nucleus pulposus-derived mesenchymal stem cells is detected, the role of MANF in
acidosis is explored by knocking down or overexpressing MANF, downstream target points are identified by using
transcriptome sequencing, and the treatment potential of NP-MSCs overexpressing MANF is evaluated by using a rat puncture model. The application explores a new
pathological mechanism that an acidic environment in an
intervertebral disc influences the repair capacity of endogenous
nucleus pulposus-derived mesenchymal stem cells, and further causes the progression of intervertebral
disc degeneration, and reveals the protection role of MANF in intervertebral
disc degeneration, thereby opening up a new idea for the treatment of intervertebral
disc degeneration.
