Methods of evaluating protein formulation stability and surfactant-stabilized insulin formulations derived therefrom

Embodiments of the invention are directed to a method of estimating the physical stability of a protein formulation. A particular embodiment of the invention places the protein formulation under an agitational stress that causes the protein to aggregate at an accelerated rate. In one embodiment, the change in protein aggregation is monitored spectroscopically using Thioflavin-T. Embodiments of the invention then utilize a survival curve analysis to ascertain the relative physical stability of the different protein formulations under study. This method was used to develop novel surfactant-stabilized insulin formulations in a rapid, cost efficient manner, thus illustrating the utility of the inventive method to the discovery and development of pharmaceutical protein formulations.

A cylinder of flowing air directed around the circumference of the lens is effective to maintain the lens in an unfolded position as the lens is being released into the processing tray. A second passage extends through one of the poles to an opening in the pole face which is covered by the armature when the latter occupies one home position but is uncovered when the armature occupies its other home position. A third fluid passage extends through and leads to a second opening in another pole, which is covered when the armature occupies its said other home position. Passages in the poles house energy storing springs each of which is compressed as the armature approaches the pole. A push rod can extend through a passage in one of the poles for conveying armature movement externally of the device.

The pump pulses are coupled through lenses to the AOM gating device, which is synchronized to the pump laser and is operable to gate the pump pulses to a reduced repetition rate Rr=Rf / N, where Rf is the pump laser fundamental frequency. The pulses from the AOM are injected via optics into the PCF. Propagation through the PCF causes the pulses to broaden spectrally to produce optical supercontinuum pulses. An optical pulse source that further includes an acousto-optical tunable filter (AOTF) operable to convert the optical supercontinuum pulses into wavelength variable output pulses is also provided. A method of scaling the energy of the optical supercontinuum pulses is also disclosed.

Reduction of fouling in heat exchangers

A method for reducing the formation of deposits on the inner walls of a tubular heat exchanger through which a petroleum-based liquid is flowing comprises applying one of fluid pressure pulsations to the liquid flowing through the tubes of the exchanger and vibration to the heat exchanger to effect a reduction of the viscous boundary layer adjacent the inner walls of the tubular heat exchange surfaces. Reduction of the viscous boundary layer at the tube walls not only reduces the incidence of fouling with its consequential beneficial effect on equipment life but it also has the desirable effect of promoting heat transfer from the tube wall to the liquid in the tubes. Fouling and corrosion are further reduced by the use of a coating on the inner wall surfaces of the exchanger tubes.

The bulk glass substrate is preferably made from a soft silica-based material having an annealing point less than about 1380°K. A pulsed laser beam is focused within the substrate while the focus is translated relative to the substrate along a scan path at a scan speed effective to induce an increase in the refractive index of the material along the scan path. Substantially no laser-induced physical damage of the material is incurred along the scan path. Various optical devices can be made using this method.

Furthermore, during pattern formation, the copolymer develops relatively low surface energy at an interface with the surface of a mold, as compared to conventional polymeric materials, and preferentially adheres to the substrate (14) rather than the mold, which provides for relatively easy mold release. The presence of the organosilicone component in the copolymer also allows the resist film (12) to exhibit excellent resistance to oxygen plasma etching.
