Genistin is an isoflavone found in a number of dietary plants like soy and kudzu. It was first isolated in 1931 from the 90% methanol extract of a soybean meal, when it was found that hydrolysis with hydrochloric acid produced 1 mole each of genistein and glucose. Chemically it is the 7-O-beta-D-glucoside form of genistein and is the predominant form of the isoflavone naturally occurring in plants. In fact, studies in the 1970s revealed that 99% of the isoflavonoid compounds in soy are present as their glucosides. The glucosides are converted by digestive enzymes in the digestive system to exert their biological effects. Genistin is also converted to a more familiar genistein, thus, the biological activities including antiatherosclerotic, estrogenic and anticancer effects are analogous.

The invention discloses a production method of soy isoflavone with high content of genistin. The production method comprises the following steps: crushing a raw material namely soybean meal, leaching by using ethanol, filtering and screening by using a screening machine, distilling to recover ethanol, precipitating isoflavone by using n-butyl alcohol, filtering to separate out a precipitation solid-phase material generated by the isoflavone in an n-butyl alcohol solution, performing washing separation and purification for the first time, and performing washing separation and purification for the second time. The production method disclosed by the invention has the positive effects that the purity of the obtained soy isoflavone is more than or equal to 90% and the content of genistin is more than or equal to 80%. The content of genistin is improved, and the soy isoflavone with high content of genistin has exact curative effects of improving the development of female secondary sex characteristics, preventing and curing climacteric syndromes, preventing female senile dementia and height atrophy, preventing and curing cancers related to estrogen secretion and the like. Therefore, the misgiving of consumer groups on the medical healthcare function of soy isoflavone is eliminated, and the production method disclosed by the invention can play an active promotion effect on the further development of the soy isoflavone industry.

Method for simultaneously extracting various kinds of pueraria isoflavones glycoside from radix puerariae crude extract

The invention provides a method for simultaneously extracting various kinds of pueraria isoflavones glycoside from a radix puerariae crude extract. The method comprises the following steps that (1) ice water is added into the radix puerariae crude extract, stirred and centrifugally filtered, and a filter cake is obtained; (2) the filter cake is dispersed by using lower alcohol, then a filling agent is added and stirred to be uniform, vacuum drying is conducted, and a stirring sample filling agent is obtained; (3) a fresh filling agent is added into a chromatographic column, and then the stirring sample filling agent is filled onto the chromatographic column; and (4) an eluent with the concentration being incremental is utilized to conduct gradient elution, eluant is collected and subjectedto vacuum concentration, cooling, stirring crystallizing, centrifugal filtration and drying, and genistin, daidzin and puerarin are obtained respectively. The content of genistin obtained by adoptingthe method is as high as 98.7%, and the yield is as high as 92%; the content of daidzin is as high as 98.5%, and the yield is 92%; and the content of puerarin is as high as 99.9%, and the yield is 97%. The method is easy to operate, low in cost, environmentally friendly, and suitable for industrialized production.
