This invention discloses a novel reverse transcriptase and its applications. The amino acid sequence of the reverse transcriptase is selected from any of the amino acid sequences shown in SEQ ID NO: 1 to SEQ ID NO: 19, or, obtained by mutating one or more amino acids of any of the above sequences, resulting in an amino acid sequence with at least 85%, 90%, or 95% identity to the original sequence and possessing reverse transcription activity. The genome-guided editing system containing the reverse transcriptase of this invention further includes a nucleic acid-programmable DNA-binding protein and pegRNA. Both the reverse transcriptase and the genome-guided editing system containing it can be used to target and modify the plant cell genome or to cultivate plants with genome modifications. This invention effectively overcomes the dependence of existing guided editing systems on single-structure chassis enzymes, thereby providing a rich reserve of underlying components for crop molecular design breeding.
