The invention relates to the field of magnetic resonance imaging and fluorescence imaging contrast agents. With the contrast agent provided by the invention, problems of existing contrast agents of low relaxation efficiency, high toxicity, low tissue organ selectivity and fast in-vivo metabolism can be solved. According to the invention, ethylenediaminetetraacetic acid (EDTA) or diethylene triaminepentaacetic acid (DTPA) are connected to folic-acid-modified bovine serum albumin fluorogold nano-clusters through amide bonds; and EDTA or DTPA is respectively coordinated with paramagnetic metal ions, such that the complex is obtained. With the contrast agent provided by the invention, the relaxation efficiency can reach 23.7mM<-1>*s<-1>. The contrast agent has an advantage of low toxicity. According to the invention, high spatial resolution of magnetic resonance imaging and high sensitivity of fluorescence imaging are combined. Therefore, the contrast agent provided by the invention can be used in magnetic resonance imaging and fluorescence imaging.

The method provided by the invention ahs the characteristics of simple process, low cost and easiness for large-scale production. The invention relates to a magnetic resonance contrast agent using a human endogenous protein as a chelating agent and preparation thereof. The contrast agent comprises the chelating agent and paramagnetic metal ions coordinated with the chelating agent, the chelating agent is the human endogenous protein, and the human endogenous protein includes aspartic acid and glutamic acid; during the preparation, firstly the human endogenous protein is added to water, the human endogenous protein is dissolved by stirring, and then trisodium citrate dihydrate is added and stirred evenly to obtain a mixedsolution; paramagnetic metal salt is added and stirred to coordinate the paramagnetic metal ions with the chelating agent so as to obtain a reaction solution, finally the pH of the reaction solutionis adjusted to 5.0-7.4, and then the reaction solution is separated and dried. Compared with the prior art, the contrast agent is formed by the direct chelation of the human endogenous protein and theparamagnetic metal ions, and traditional micro-molecule chelating agents are not needed; the contrast agent has simple synthesis process, mild reaction conditions and good environmental compatibility, reduces energy consumption, and is easy in mass production.

The MRI macromolecular contrast agent can be prepared according to the following steps of: a) dissolving a fullerene carboxylic acid derivative in a 2-(N-morpholino) ethanesulfonic acid (MES) buffer solution, adding 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS), and reacting at room temperature to obtain a fullerene carboxylic acid active ester solution; and b) adding gadolinium diethylenetriamine pentaacetic acid human serum albumin (Gd-DTPA-HAS) into the fullerene carboxylic acid active ester solution, stirring, reacting, and then obtaining the MRI contrast agent. The relaxation rate of the Gd-DTPA-HAS-fullerene macromolecular contrast agent is obviously improved under the condition of low field of 0.5T, and obvious tissue contrast ratio can be obtained under the condition of reducing the dosage of the contrast agent; and most importantly, the MRI contrast agent can be used as an MRI molecular probe which can be used for exploring body distribution andmetabolism behavior of nanometer materials such as functionalized fullerene derivatives.
