This invention belongs to the field of
medicinal chemistry, specifically relating to a mitochondrial-dequinoline
chloride-
hyaluronic acid compound, its preparation method, and its application. The rotenol-dequinoline
chloride-
hyaluronic acid compound has the following
structural formula, where n is 2. Based on the anticancer activity of
rotenone, this invention utilizes reduction, esterification, and two
amide reactions to successfully link rotenol, dequinoline
chloride, and
hyaluronic acid via
amide bonds. Subsequently, these components are successfully self-assembled into spherical nanoparticles using
ultrasound, constructing a rotenol-dequinoline chloride-hyaluronic acid nanosystem for
tumor therapy. After targeted delivery of these nanoparticles to
tumor cells, both rotenol and dequinoline chloride synergistically act on the mitochondria, forming a dual-
drug delivery system that generates a large amount of ROS, thereby inducing
apoptosis and effectively inhibiting
tumor growth and migration.
