Method of reducing content of amylase in hybrid rice

The invention discloses a method of reducing the content of amylase in hybrid rice and belongs to the technical field of rice genetic breeding. In the method, an original hybrid rice combination: one of a parent of a non-glutinous sterile line / a non-glutinous restorer is modified into a glutinous parent. The hybrid rice is prepared through a hybridization method of a glutinous sterile line / non-glutinous restorer or a non-glutinous sterile line / a glutinous restorer to obtain a hybrid rice variety in which the content of the amylase is lower than that of the original hybrid rice. In the invention, by means of a genetic effect and a genetic dosage effect of a glutinous gene wx, a near-isogenic line combination is reduced in the content of the amylase. In addition, a growth period, a plant and leaf form, panicle traits, floral organ traits, yield traits, disease resistant performance, adaptability and the like of the near-isogenic line combination are free of significant different when the near-isogenic line combination is compared with the original hybrid rice combination. Through quality analysis and taste evaluation, a new hybrid rice variety being appropriate in the content of the amylase and having the taste and the quality which are suitable for people in different region, thereby achieving an object of genetically improving a poor taste and a poor quality due to a high amylase content in the hybrid rice in the prior art.

Method for modifying corn starch with different amylose contents by air atmospheric pressure plasma jet

The invention discloses a method for modifying corn starch with different amylose contents by air atmospheric pressure plasma jet. The method comprises preparing waxy corn starch, common corn starch and high amylase corn starch into starch emulsions having a mass concentration of 5-15%, directly treating the starch emulsions with an air atmospheric pressure plasma jet device for 1-7 minutes, centrifuging the starch emulsions to obtain precipitates, washing the precipitates for 3 times with absolute ethanol, and performing vacuum freeze drying to obtain modified starch. The method uses the airatmospheric pressure plasma jet technology to treat the three corn starch emulsions, and introduces some oxygen-containing polar groups into the corn starch for hydrophilicity improvement, mainly reflected in increase of water bonding ability and water expansion volumes, of the starch so as to further improve the practical application performance of the three kinds of corn starch. The method can also provide a new direction for starch modification for scholars who study the direction in the future, and provides more theoretical supports for the starch research field. The modified starch produced by the method can also be better applied to convenience food.

Screening method for mutated strains capable of realizing high yield of amylase

The invention relates to a screening method for mutated strains capable of realizing high yield of amylase. The method comprises a step of screening of mutated strains, i.e., screening of first-generation strains having undergone mutation; the step comprises a substep of primary screening and a substep of secondary screening; the secondary screening is carried out in a manner the same as the manner used in primary screening so as to obtain second-generation mutated starting strains; then mutation and screening are carried out again; and the screening method for the generation II and generation III to generation n is same, and mutation and screening are carried out until high-amylase-yield stains meeting requirements are bred. With the screening method, the starting strains having undergone mutation treatment undergoes primary screening and secondary screening again so as to obtain the second-generation starting strains; the screening method is simple and easily practicable, and can obtain the high-amylase-yield stains in a short time; the whole screening process is low in investment and high in targeting performance; and orientated production of the amylase strains with high enzyme activity can be realized, and the method has large-scale promotion value.
