This invention relates to the field of
heme purification technology, and more particularly to a method for extracting and purifying
heme chloride. The method includes: taking fresh animal blood, centrifuging to collect the lower
red blood cell precipitate, adding
physiological saline, mixing, and washing; adding a 1-10%
ethanol solution, mixing, and ultrasonically disrupting at 300-800W for 15-60 seconds to obtain a
red blood cell lysate; adding an acidic
acetone solution to the
red blood cell lysate, mixing, and extracting at
room temperature, centrifuging to collect the supernatant to obtain an acidic
acetone extract; slowly adding NaOH solution to adjust the pH; adding
sodium acetate, stirring, allowing to stand, collecting the precipitate, washing, and
drying to obtain a crude product; packing a
silica gel column; loading the crude product onto an eluent, eluting with an eluent and collecting the eluent fraction containing the target product; the eluent includes
dichloromethane,
methanol, and
formic acid; and removing the mobile phase from the eluent fraction. The advantages are: simple process, low cost,
environmentally friendly, and the ability to obtain a high-purity product.
