heme extraction and purification method. Comprising the following steps: taking fresh animal blood, centrifugally collecting lower
red blood cell precipitate, adding normal
saline, uniformly mixing and washing; adding an
ethanol solution with the concentration of 1-10%, uniformly mixing, and ultrasonically crushing for 15-60 seconds under the power of 300-800W to obtain erythrocyte crushing liquid; adding an acidic
acetone solution into the erythrocyte crushing liquid, uniformly mixing, extracting at normal temperature, centrifuging, and collecting supernate to obtain an acidic
acetone extracting solution; slowly adding a NaOH solution to adjust the pH value; adding
sodium acetate, stirring, standing, taking precipitate, washing and
drying to obtain a crude product; filling a
silica gel column; loading the crude product, eluting with an eluent, and collecting an
elution component containing a target product; the eluent comprises
dichloromethane,
methanol and
formic acid; and removing the mobile phase in the
elution component. The method has the advantages of simple process, low cost, environmental friendliness and capability of obtaining a high-purity product.
