rootstock by overexpressing MiNAC2a

cotyledon of aseptic
seedling from macadamia seed, inducing
callus by using the
cotyledon of the aseptic
seedling, constructing an overexpression vector according to the obtained MiNAC2a
gene sequence, transferring the vector into
agrobacterium, mediating the
agrobacterium to transfer into macadamia
callus by injection method, culturing positive
callus to grow adventitious buds and adventitious roots, extracting
DNA of the
seedling to verify whether the positive
plant is true or not by detecting bands, hardening the seedling,
transplanting the seedling to a field, obtaining transgenic drought-resistant
rootstock resources, strengthening the
root system of the transgenic seedling, and improving the
drought resistance of the
rootstock. The technology uses
transgenic technology to cross boundaries, break through
reproductive isolation, and broaden the
gene resource
library, and provides a
train of thought for precisely introducing key drought-
resistant genes in
bacteria, fungi and even other drought-resistant plants into macadamia trees.
