gene engineering bacteria producing
glucose oxidase as a starting strain, adds a
fermentation medium in a fermentation tank, adds PTM1 salt after sterilization and temperature reduction, and then inoculates seed liquid for primary fermentation. When the concentration of
reducing sugar in the fermentation liquid is reduced to below 0.5%, glucose solution and PTM1 salt are added for secondary fermentation. When the concentration of
reducing sugar in the fermentation liquid is reduced to below 0.5% again, a
mixed solution of
glycerol,
methanol and water and PTM1 salt are added for
mixed feeding fermentation. The
mixed solution of
methanol and PTM1 salt is added in the fermentation liquid for induction fermentation. The
enzyme activity reaches 11309 U / mL after 192 h of fermentation. In the fermentation method, the dynamic strategy of multi-stage
carbon source control, especially the
mixed feeding stage and the induction fermentation of
methanol addition, further realizes the
cell density and long-
time efficient induction expression, so that high-yield
glucose oxidase is obtained.
