The invention belongs to the field of bioanalytical
chemistry and
drug metabolism research, and discloses a
typing quantitative detection method for different forms of iron in serum, which comprises the following steps: S1, mixing a to-be-detected serum sample with an inorganic
chlorine salt solution, placing in an
ultrafiltration centrifuge tube, standing, centrifuging, and taking filtrate as non-
transferrin binding iron NTBI; s2, a weak acid solution of inorganic
chlorine salt is added into the filter residues obtained in S1,
centrifugation is conducted, and filtrate serves as
transferrin binding iron TBI; s3, a
mixed solution of a
reducing agent solution and an inorganic
chlorine salt solution is added into the filter residues obtained in the S2,
centrifugation is conducted, and filtrate serves as
drug binding iron DI; and S4, respectively measuring
absorbance of the NTBI, TBI and DI parts by adopting a spectrophotometric method, and converting the concentration by combining a
standard curve, so as to obtain the content of the NTBI, TBI and DI in the serum sample to be detected. The method realizes high-selectivity separation and accurate quantification of three different forms of iron, and is suitable for intravenous
iron agent in-vivo behavior research and analysis of iron
metabolism related mechanisms.
