The invention discloses application of an ethylene responsive factor gene OsERF3 in regulating and controlling the development of rice roots. By using a yeast two-hybrid method, an ethylene responsive factor OsERF3 interacted with a published OsWOX11 protein is obtained by screening. By using an overexpression and small artificial interference RNA method, transformed rice corresponding to a gene is obtained, and a situation that the length of a main root and the number of crown roots of a plant subjected to gene overexpression are obviously increased in comparison with those of wild compared plants, and the length increasing of the main root is caused by the length increasing of cells in a maturation zone of a root tip, the length of the main root and the number of crown roots of a plant subjected to small artificial interference RNA transgenosis are obviously reduced in comparison with those of wild compared plants, and the length reducing of the main root is caused by the length reducing of cells in a meristematic zone and a maturation zone. Results show that the gene has a function of promoting the development of rice roots. A result of fusing the promoter of the gene with a report gene beta-glucuronidase shows that the gene is expressed in a meristematic zone of a root tip.

The invention provides a culture method for in-vitro human oocyte preantral follicles. The culture method is characterized by comprising the following specific steps of: 1) unfreezing the in-vitro human oocytes placed in frozen protecting solution containing cane sugar and propylene glycol and subjected to liquid nitrogen low-temperature storage, and then reviving and culturing; and 2) separating and extracting the in-vitro human oocytes obtained by reviving and culturing, then placing the in-vitro human oocytes in nutrient solution containing follicle-stimulating hormone (FSH), and performing preantral culture, wherein the culture for in-vitro human oocyte preantral follicles needs the support of FSH, and FSH can be used for promoting the growth of the follicles, increasing the survival time of the follicles, promoting the proliferation of granular cells, and promoting the maturation of the oocytes, as well as is an important factor for regulating and controlling the growth and development of the follicles; and trichostatin A can be used for effectively inhibiting the proliferation of tumour cells, regulating and controlling the progress of a cell cycle and then inducing the apoptosis of tumour cells, thus ensuring the survival quality of the cells. The method has important significance on researches in the aspect of test-tube babies.

Virus-free potato cutting seedling planting method

The invention discloses a virus-free potato cutting seedling planting method. The method at least comprises steps as follows: 1) a 20 cm thick substrate is paved with river sand with the size of 0.5-1mm on a seedling bed, virus-free potato cutting seedlings are transplanted, and root systems of the virus-free potato cutting seedlings are buried into the substrate; 2) after transplantation, the cutting seedlings are covered with a plastic film and a sunshade gauze, the plastic film is removed step by step after the seedlings recover for 7-10 days and the cutting seedlings grow new roots in thesubstrate, and the sunshade gauze is removed after 30-40 days of transplantation; 3) water-soluble mixed fertilizer is applied after 9-11 days of transplantation; and 4) after the potato fruiting period begins, the maximum water-holding capacity of the substrate is controlled at 75%-80%, the temperature of the growth environment is controlled at 17 DEGC-25 DEGC, and diseases, insect pest and weeds are controlled on the potatoes in the whole growth process. According to the method, the river sand is paved on the seedling bed to serve as the substrate, the roots of the cutting seedlings are buried in the substrate layer, and virus infection in the growth process of the cutting seedlings is effectively reduced.
