powder is used as a
raw material to extract
total protein,
salting-out crude
protein is obtained through
salting-out, and high-purity acidic
protein is obtained through purification; the method comprises the following steps: by taking
salting-out crude
protein as a substrate, carrying out step-by-step enzymolysis (40DEG C, pH 7.5 and 1h respectively) on flavourzyme (3000U / g) and
trypsin (1000U / g), carrying out 10kDa and 3kDa
ultrafiltration grading on enzymatic
hydrolysate, and determining that a component with acidic protein less than 3kDa is an optimal component (Ka = 38.8067 + / -1.2, and a threshold value is 0.176 mg / mL) by combining an
electronic tongue, sensory evaluation and a delicate
flavor threshold value.
Peptide fragments are identified through LC-MS / MS, 30 non-toxic and non-
sensitization potential
umami peptides are obtained through
virtual screening, and molecular docking verifies that the
umami peptides are stably combined with a T1R1-VFT
receptor. The process has synergistic interaction, the product is natural and safe, and high-quality raw materials and technical schemes are provided for development of natural seasonings. The abstract drawing is figure 3.
