The invention relates to an absorbable magnesium alloy staple having antibiosis and anti-inflammation functions, and a making method thereof. The staple is composed of a pure magnesium or magnesium alloy wire core with the surface containing a porous ceramic layer, an intermediate functional layer loaded with anti-inflammation medicines and copper ions with gelatin as a carrier, and a degradable high-molecular external surface layer. The copper ions and the anti-inflammation medicines are effectively loaded on the surface of the wire with gelatin as the carrier by adopting an electrophoresis-electrodeposition process, and the external surface layer of the staple is a degradable high-molecular layer having a protection effect. The staple can slowly release the copper ions having a sterilizing effect and the anti-inflammation medicines having an anti-inflammation effect in the use process. Additionally, the synergistic effect between the intermediate functional layer with the gelatin as the carrier and the external degradable high-molecular layer realizes the effective control of the medicine release rate and the staple degradation speed. The invention discloses a method for preparing a matrix-induced autologous bone mesenchymal stem cell composite scaffold. The method specifically comprises the steps of preparing bone marrow suspension; taking peripheral blood of a patient and putting the peripheral blood of the patient in a blood bag; preparing autoserum; obtaining target cells; adding 15 ml DMEM high glucose culture medium to the target cells, inoculating a T25 cell culture bottle with the target cells, and placing the T25 cell culture bottle in an incubator for culture; rinsing the cells twice with PBS solution; adding the DMEM high glucose culture medium into the culture bottle dropwise to stop digestion; obtaining cleaned sediment; inoculating the cell culture bottle with the cells for continuous culture; conducting passage three times; adding trypsin-EDTA solution into the culture bottle; washing the cells three times with PBS; conducting cell resuspension; placing a sterile pig source I/III type collagen membrane in a sterile plastic culture dish, and soaking the membrane in the DMEM high glucose culture medium for one night; and dropping cell suspension onto the pig source I/III type collagen membrane, and conducting culture for 3 days. The method has the advantages of being high in treatment precision and the like.

Preparation method for collagen membrane inoculated with autologous mesenchymal stem cells

The invention discloses a preparation method for a collagen membrane inoculated with autologous mesenchymal stem cells. The preparation method comprises the following concrete steps: carrying out uniform mixing so as to obtain a marrow suspension; preparing autologous serum; adding the marrow suspension into a centrifuge tube containing a cell separating medium and carrying out gradient centrifugation to obtain a sediment which is target cells; adding the target cells into 15 ml of a DMEM high-glucose medium containing FGF-2, the autologous serum and gentamycin, carrying out culturing in an incubator and replacing the DMEM high-glucose medium once every three days; after cell culture for 3 to 4 weeks, carrying out passage three times; subjecting cells to resuspension with normal saline; slowly adding the cell suspension onto the rough surface of a prepared pig-sourced I/III type collagen membrane with a plastic sleeve drop by drop; and allowing the collagen membrane to adsorb cells for 10 to 15 min so as to obtain the collagen membrane inoculated with the autologous mesenchymal stem cells. The collagen membrane inoculated with autologous mesenchymal stem cells prepared in the invention has the advantages of good treatment effect, convenience in treatment, low cost, etc.
