The invention provides a method for increasing the cross compatibility and the seed setting rate of tobacco and N.alata, and belongs to the technical field of agricultural
biology. The method includes the steps of starting hanging female parent flowers, wherein the hanging time is the flowering
start time, and the 72nd hour, the 96th hour and the 120th hour after hanging are the aging stages after the flowering
start time; pollinating female parent stigmas with male parent mature
pollen during the three time periods, conducting sampling 48 hours after
pollination, counting the number of
pollen tubes and the seed setting rate through
callose fluorescence, and comparing influences of
pollination on the cross compatibility; selecting flowers, at the flowering
start time, of female parents, stripping pistils, clipping stigmas which are smashed into
powder after being naturally aired indoors, and mixing the male parent mature
pollen with sigma
powder according to the weight ratio between 1 to 5 and 1 to 50; pollinating the flowers with mixed pollen 120 hours after the flowering start time of the female parents, conducting sampling 48 hours after
pollination, counting the number of pollen tubes and the seed setting rate through
callose fluorescence, and comparing influences of pollination on the cross compatibility; cultivating tobacco as the female parents, and planting N.alata as the male parents. The method has the advantages that operation is simple, cost is low, popularization is easy, and reference is provided for obtaining of
plant inter-species coeno-species.
