The invention discloses a new medical application of loganin, and in particular relates to an application of the loganin in preparation of medicaments for treating diabetes, wherein the loganin is extracted from dogwood. The method for extracting the loganin comprises the following steps of: (1) performing water extraction and alcohol precipitation on dogwood coarse powder to obtain a dogwood water extraction and alcohol precipitation extracting solution; (2) loading the dogwood water extraction and alcohol precipitation extracting solution into a macroporous adsorption resin column, and performing enrichment to obtain dogwood iridoid glycosides; (3) performing chromatographic separation by using a silica gel column to obtain a loganin flow component; and (4) purifying to obtain a loganin monomeric compound. Pharmacodynamics experimental results prove that the loganin has the functions of reducing the content of blood sugar, serum AGEs and urokinase protein, raising the serum insulin level, relieving pathologic lesions of pancreas, kidney, liver, heart, testis and the like, and obviously improving a 'three more one less' symptom. Therefore, the loganin can be used for preparing the medicaments for treating the diabetes.

Novel method for simultaneously measuring contents of multiple active ingredients of dogwood

The invention discloses a novel method for simultaneously measuring contents of multiple active ingredients of dogwood. The method is used for simultaneously measuring the contents of seven ingredients, namely loganin, gallic acid, 5-HMF (hydroxymethyl furfural), protocatechuic acid, morroniside, sweroside and cor-nuside, in dogwood medicinal materials from four main producing areas by utilizing a high performance liquid diode array detection method. The method comprises the steps of: carrying out gradient elution at the flow speed of 1.0 mL.min-1 by utilizing a Phenoemenex C18 polar chromatographic column and taking acetonitrile and 0.1% methanoic acid solution as mobile phases, wherein the column temperature is 30 DEG C, the detection wavelengths are 220 nm, 240 nm, 260 nm, 265 nm and 280 nm. The contents of the loganin, the gallic acid, the 5-HMF, the protocatechuic acid, the morroniside, the sweroside and the cor-nuside in each sample are respectively 0.22-0.800 mg/g, 0.002-0.024 mg/g, 0.069-3.775 mg/g, 8.192-28.111 mg/g, 0.048-0.598 mg/g, 1.228-9.311 mg/g and 1.450-8.381 mg/g. The method is simple and accurate, is good in reproducibility and is capable of providing bases for comprehensively evaluating and controlling the quality of dogwood decoction pieces.

The invention provides a multi-index fast detection method of dogwood medicinal materials. The multi-index fast detection method comprises the following steps of: acquiring the dogwood medicinal materials; determining the contents of morroniside and loganin in the dogwood by using a high-performance liquid chromatography, determining the content of water by using a drying method, and determining the content of extract by using a cold-extraction method; collecting near-infrared spectrum data of dogwood medicinal material powder; preprocessing spectrum at the modeling waveband of the near-infrared spectrum; establishing a fast detection model for the water, the extract and the morroniside and the loganin in the dogwood medicinal materials; and using the established model for fast determination of the water, the extract and the morroniside and the loganin in the dogwood medicinal materials. The multi-index fast detection method provided by the invention has the advantages that a near-infrared spectrum technology is introduced as the fast detection method of the contents of the water, the extract, the morroniside and the loganin in the dogwood medicinal materials; and compared with the traditional method, the established analysis method has the advantages that whether the quality of the medicinal materials is qualified can be fast judged, whether the medicinal materials can enter the following production-process links can be determined, the requirements of fastness and high efficiency in production can be met, and the application prospect for site medicinal-material screening and comprehensive quality evaluation is achieved.

The method is simple and easy in operation and mild in conditions and is suitable for simultaneously preparing and producing morroniside of which the purity is greater than or equal to 98% and loganin of which the purity is greater than or equal to 98% in a large scale from the dogwood extract.
