This invention provides an oral hydrogel
microsphere for targeted intestinal inhibition of
gout, its preparation method, and its application, belonging to the field of
biomedical technology. The preparation method of the oral hydrogel
microsphere of this invention includes the following steps: (1) dissolving uricase and
dopamine in
sodium alginate solution, controlling the concentration of uricase to 10-20 U / mg and the concentration of
dopamine to 0.01-0.1 g / ml, and then dispersing by
ultrasound; (2) placing the solution obtained in step (1) in a microfluidic device, and using gas microfluidic technology, shearing the solution into uniform droplets under the action of
nitrogen flow; (3) dropping the droplets obtained in step (2) into
calcium chloride solution, and cross-linking to form the oral hydrogel
microsphere for targeted intestinal inhibition of
gout. This invention successfully constructs intestinal "
bursting" oral
hydrogel microspheres loaded with uricase, effectively immobilizing uricase in the
intestinal mucosa, thereby inhibiting
gout for the first time by excreting free
uric acid through the intestine. The microspheres have high uricase activity and good
therapeutic effect on gout.
