The invention relates to the microbial fermentation field and discloses a method of utilizing dendrobium candidum slag for cultivating ganoderma through solid fermentation. The method utilizes dendrobium slag as medium to perform the solid fermentation of the ganoderma and is achieved through the following steps of (1) slant cultivation, (2) seed cultivation and (3) solid fermentation cultivation. The cultured seeds are inoculated into a solid fermentation culture medium at an inoculation size of 10%. The dry materials of the solid fermentation culture medium are composed of 98% of the dendrobium slag, 1.9% of bran and 0.1% of potassium hydrogen phosphate and the weight ratio of the dry materials and water is 1: (1-2.3). The ganoderma is cultivated for 7-20 days at a natural pH value in a temperature condition of 22-32 DEG C. According to the method of utilizing the dendrobium candidum slag for cultivating ganoderma through the solid fermentation, the cellulose in the dendrobium slag is decomposed and utilized by the ganoderma through the fermentation and the biotransformation of the ganoderma to generate high-efficiency bioactive materials such as ganoderma polysaccharide and ganoderma acid, and the deep-processing waste of dendrobium products is converted into high value-added products, so that the all-around comprehensive utilization of dendrobium raw materials is achieved.

The method particularly comprises the following steps of: (1) pulverizing fresh or dry ganoderma lucidum fruiting bodies; (2) establishing an ethanol/ammonium sulfate aqueous two-phase system; (3) performing extraction separation: adding ganoderma lucidum powder into the ethanol/ammonium sulfate aqueous two-phase system, shaking up the mixture by vibration or uniformly mixing the mixture by ultrasonic sound, and standing the mixture for phase splitting or centrifugally splitting the phase to obtain a top phase and a bottom phase; (4) concentrating the top phase under reduced pressure and freeze-drying the top phase to obtain the ganodenic acid; and (5) dialyzing the bottom phase to obtain permeate and intercept fluid, performing alcohol precipitation on the intercept fluid with 80 percent ethanol and centrifuging to obtain a precipitate and supernatant, and free-drying the precipitate to obtain the ganoderma lucidum polysaccharide. The method has the advantages of simple process, low equipment requirement, low cost, no pollution, suitability for batch production and capability of realizing synchronous extraction of the ganodenic acid and the ganoderma lucidum polysaccharide.

The invention relates to a quality analysis analysis method of medicines or health care products, and specifically relates to a quality controlling method of a ganoderma aqueous extract. For controlling the product quality with high specificity, according to the invention, crude polysaccharide contents and a sum of ganoderma acid A, B, and C2 contents are adopted as quality controlling indexes of the ganoderma aqueous extract. According to the invention, ganoderma crude polysaccharide is extracted with a water extraction and alcohol precipitation method; polysaccharide with a glucan structure is precipitated by using a copper reagent; and the content of the polysaccharide is determined by using an ultraviolet spectrophotometry method. Therefore, the influences of auxiliary materials on determination are eliminated, and a basis is provided for quality controlling of the ganoderma aqueous extract. The contents of the characteristic components ganoderma acid A, B, and C2 in ganoderma lucidum, ganoderma sinnese, and ganoderma tsugae are relatively high. Compared to a currently commonly-used colorimetric method for determining total triterpenoid contents, the specificity of the method provided by the invention is high. With the method, purposes of authentication and quality control can be achieved.
