Method for screening hypha-state candida albicans virulence related genes

The invention discloses a method for screening hypha-state candida albicans virulence related genes. The method comprises the following steps of: firstly, obtaining the candida albicans growing in a hypha phase by adopting a hypha induction experiment, then, analyzing the transcriptome difference of the candida before and after hypha induction by using a transcriptome sequencing method (RNA-seq), screening virulence related genes, and then, detecting and analyzing respective damage conditions of different host cells infected by the candida albicans by adopting a gram staining method and a cell proliferation rate; and finally, analyzing the difference of virulence gene expression after different types of host cells are infected by the candida by using a reverse transcription-fluorescent quantitative PCR method (qRT-PCR). According to the method, the associated relationship between hypha formation and some virulence factors of the candida can be defined; potential virulence related genes which are differentially activated when different cells are infected by the candida are screened out; and evidence is provided for illuminating different molecular mechanisms of candida infected hosts and screening high-specificity and high-reliability candida virulence genes for diagnosis.

Preparation and application of ATM fusion protein for preventing infection of S. aureus and C. albicans

The invention relates to ATM fusion protein. 9 amino acids of the T-cell epitope of C. albicans Als3 are connected to the front of the amino acid sequence of S. aureus MntC fusion protein through a Linker, and the amino acid sequence of the fusion protein is shown in SEQ ID No. 1. In the invention, the T-cell epitope of Als3 is fused with the MntC protein so as to prepare the recombinant ATM fusion protein, and immunogenicity study is conducted on the ATM fusion protein. It is verified through results that organisms are induced to produce better immune response and resistance to S. aureus infection and a better immune protection effect after the T-cell epitope of Als3 is fused with the MntC protein, and MntC-induced effects of immune-protection and resistance to S. aureus infection are enhanced significantly through the T-cell epitope of Als3; and meanwhile, the ATM fusion protein also has certain effect of resistance to C. albicans infection, and is ideal candidate fusion protein forpreparation of reagents with resistance to S. aureus infection and C. albicans infection, and the fusion protein has better immunogenicity, and is capable of inducing the organisms to produce good effects of immune protection.
